人脐带间充质干细胞的培养鉴定及其向神经元细胞分化的研究

摘要: [摘要]目的 研究脐带间充质干细胞(UC-MSCs)在体外分离、培养、扩增和鉴定以及向神经元分化的方法.方法 30例符合入选标准的脐带采用贴壁法分离得到UC-MSCs,选用20% FBS血清培养液进行原代培养,再选用10% FBS扩增培养液进行一般传代.用神经分化培养基诱导UC-MSCs向神经细胞分化,在荧光显微镜下观察细胞形态, 透射电镜观察UC-MSCs超微结构,用流式细胞仪分析其表面特性,采用直接和间接免疫荧光法检测细胞悬浮液的表型特征,用抗人神经纤维 M、突触素、微管蛋白、半乳糖神经酰胺的单克隆抗体检测各代之间神经诱导形成率.结果 12 h后细胞开始贴壁生长,超微结构观察表现出未分化细胞的特征,CD123、CD49、CD29、CD73和CD166均为阳性,免疫荧光检测人UC-MSCs分化成神经细胞.结论 UC-MSCs长期传代培养,生物学特性稳定,具有向神经细胞分化的潜能
