人脐带间充质干细胞的培养鉴定及其向神经元细胞分化的研究

摘要: ［摘要］目的 研究脐带间充质干细胞（UC-MSCs）在体外分离、培养、扩增和鉴定以及向神经元分化的方法．方法 30例符合入选标准的脐带采用贴壁法分离得到UC-MSCs，选用20% FBS血清培养液进行原代培养，再选用10% FBS扩增培养液进行一般传代．用神经分化培养基诱导UC-MSCs向神经细胞分化，在荧光显微镜下观察细胞形态， 透射电镜观察UC-MSCs超微结构，用流式细胞仪分析其表面特性，采用直接和间接免疫荧光法检测细胞悬浮液的表型特征，用抗人神经纤维 M、突触素、微管蛋白、半乳糖神经酰胺的单克隆抗体检测各代之间神经诱导形成率．结果 12 h后细胞开始贴壁生长，超微结构观察表现出未分化细胞的特征，CD123、CD49、CD29、CD73和CD166均为阳性，免疫荧光检测人UC-MSCs分化成神经细胞．结论 UC-MSCs长期传代培养，生物学特性稳定，具有向神经细胞分化的潜能
