黄芩苷抗脑缺血小鼠学习记忆功能损伤的保护作用

摘要: 目的 探讨中药黄芩苷对脑缺血-再灌注模型小鼠学习记忆功能损伤的保护作用和机理.方法 将60只小鼠随机等分为脑缺血模型组、假手术组、黄芩苷组,采用经典Morris水迷宫法测试小鼠空间学习记忆能力,记录每天的潜伏期与游泳轨迹,同时运用蛋白印记的方法来检测小鼠纹状体中Bax 和Bcl-2蛋白水平的变化.结果 黄芩苷组小鼠水迷宫训练学习期的第4 天开始,逃避潜伏期缩短、错误次数减少,与模型组相比差异有统计学意义(P < 0.01);脑缺血模型组损伤4 d后,小鼠纹状体区神经细胞Bcl-2与Bax表达、Bax/Bcl-2比值都上调;黄芩苷组预处理可以分别下调Bax表达与Bax/Bcl-2比值,较模型组显著降低(P<0.01).结论 黄芩苷能有效预防脑缺血-再灌注致小鼠学习能力的损伤,其作用机制可能与Bax/Bcl-2改变有关.Abstract: Objective To investigate the protective effect and mechinasm of baicalin against the learning and memory capacity of mice with cerebral ischemia-reperfusion injury.Methods Sixty Kunming mice were randomly divided into three groups:control group(n=20),reperfusion group(n=20)and baicalin group(n=20).Water maze of Morris was used to detect the learning and memory capacity,and latent period and swimming locus were recorded.Western blot analysis was used to detect Bax and Bcl-2 protein expression in mice in each...
