一重和三重脑震荡大鼠脑小胶质细胞定量变化研究

摘要: 目的建立一重脑震荡(PCC)和三重脑震荡(MCC)大鼠模型,通过计数小胶质细胞(microglia,MIG)数目变化,观察PCC和MCC鼠脑的MIG病理学变化规律,以探讨MCC大鼠脑损伤的累加效应.方法用自制金属单摆式闭合性脑损伤打击装置复制PCC和MCC大鼠模型96只,随机分为PCC鼠l d、2d、4 d、8 d、16 d、24 d组和MCC鼠l d、2 d、4 d、8 d、16 d、24 d组(n=6)共十二个实验组,另设一正常对照组(n=6).运用HE组织化学染色,对中隔断面的前额叶皮质(PFC)、梨状皮质(Pir)、尾壳核(CPu)、伏隔核(AcbC)、终纹床核(BST);背侧海马断面的无颗粒细胞皮质(RSA)、海马CA1-4区、上、下齿状回、丘脑背外侧核(LD);脑干白质以及被盖区等15个脑区进行MIG计数.结果大鼠PCC和MCC后绝大多数脑区(除前额叶皮质和海马)的MIG数目均呈明显上升改变,且MCC后AcbC、Pir、CPu、BST、RST、CA3、LD和上齿状回部位的MIG数目上升变化比PCC鼠更加显著(P<0.05).结论大鼠PCC和MCC后绝大多数脑区的小胶质细胞数目均呈上升性改变,而且M...Abstract: Objective To establish Pure Cerebral Concussion(PCC)and Multiple Cerebral Concussion(MCC)rats model and observe the changes of microglias(MIG)by cell count after injury,so as to understand the additive effect mechanisms of MCC.Methods The PCC or MCC model was duplicated by using a metallic pendulum-striker concussive device.96 female and male Sprague-Dawley rats were randomly divided into twelve groups:2 d,4 d,8 d,16 d,24 d groups and MCC 1 d,2 d,4 d,8 d,16 d,24 d groups,one control group was used,AND eight...
