多聚乙酰亚胺介导CNP基因对兔髂动脉球囊成形术后内膜增生的影响

摘要: 目的观察局部转染C型利钠肽基因(CNP)对血管内膜增生的影响,探讨CNP在防治球囊成形术后再狭窄中的作用.方法构建兔髂动脉粥样硬化模型,将多聚乙酰亚胺-pcDNA3.1/hCNP包被球囊转染髂动脉,分别用RT-PCR、免疫组化、Western blot等方法检测pcDNA3.1/hCNP在动脉壁中的表达,弹力纤维染色法检测球囊成形术后内膜增生等指标.结果 pcDNA3.1/hCNP在兔髂动脉中能高效表达,转染组血管内膜增生程度明显降低.结论 CNP基因转染能明显抑制血管内膜增生,在血管再狭窄防治中可能具有应用价值.Abstract: Objective To discuss the potential role of C-type natriuretic peptide(CNP)in vascular restenosis prevention after aortic sac plasty by observating the influence of CNP on the intima hyperplasia.Methods Reconstructed plasmid pcDNA3.1/hCNP was transfected with polyethylenimine(PEI)by catheter method in iliac artery of rabbit model of atherosclerosis.The expression of pcDNA3.1/hCNP was confirmed with RT-PCR,immunohistochemistry and Western blot.The hyperplasia of intima was detected with elastic fibers stainin...
