多西紫杉醇联合伽马刀对肝癌细胞裸鼠皮下移植瘤凋亡作用的影响

  • 摘要: 目的 探讨多西紫杉醇联合伽马刀对肝癌细胞株裸鼠皮下移植瘤细胞凋亡作用的影响.方法 成功建立:SMMC-7721肝癌细胞株裸鼠皮下移植瘤模型,分2组,每组各10只裸鼠,即多西紫杉醇联合伽马刀组及肿瘤对照组.实验组先用注射多西紫杉醇处,剂量为60[μg/(0.3 m L·只)],每3天注射1次,连续注射6次,隔天伽马刀照射1次,连续伽马刀照射6次.(室温下用137Cs放射源照射局部肿瘤,照射按分割照射每次分别给予5 Gy总剂量为10 Gy).对照组注射生理盐水,剂量同前.观察2组的肿瘤生长情况.30 d后切取瘤样组织,用TUNEL法检测肿瘤细胞凋亡率.结果 实验组的肿瘤生长速率明显减慢,实验组的凋亡指数(35.03±1.51)%明显高于对照组(0.87±0.39)%,两者间差异有统计学意义(P<0.05).结论 多西紫杉醇联合伽马刀具有促进肿瘤细胞凋亡进而抑制肝细胞癌裸鼠移植瘤生长作用.
