miRNA-192在人体手术切除肺癌组织中的表达及临床意义研究

摘要: [摘要]目的 发掘与肺癌相关的特异性miRNA用于肺癌的分型分期和预后判断.方法 对47例肺癌患者手术切除的肺癌组织和癌旁正常肺组织用SYBR Green实时荧光定量PCR对miRNA-192进行定量检测,比较肺癌组织和癌旁正常肺组织miRNA-192的表达是否有差异.结果 吸烟者,每天吸烟支数>3支者,吸烟年数>2 a及饮酒者的肺癌患者肺癌组织miRNA-192表达量较不吸烟、每天吸烟支数<3支者,吸烟年数<2 a及不饮酒者高.总的47个病例miRNA-192在肺癌组织中表达量△CT值为(8.19±2.94),正常肺组织的表达量ΔCT值为(7.42±2.64),经配对t 检验,P>0.05 ,miRNA-192在肺癌组织中的表达与正常肺组织差异无统计学意义(P>0.05);经相关分析肺癌组织中的miRNA-192表达倍数与临床资料之间无相关关系.结论 miRNA-192在具有吸烟及饮酒危险因素的人群中表达增高,可认为miRNA-192在肺癌的发生中具有一定的作用,但因总的47个病例的miRNA-192在肺癌组织和正常肺组织中表达差异无统计学意义,因此还无法确定miRNA-192是否属于肺癌的癌基因或抑癌基因,miRNA-192目前还不能用于肺癌的分型分期和预后判断.

Abstract: [Abstract]Objective To explore the lung cancer related specific miRNA for judging the pathological type and disease progression.Methods The 47 samples were got by multi-stage randomized sampling. TRIzol was used to extract the total of RNA from tissue samples,real-time SYBR Green fluorescent quantitative polymerase chain reaction(PCR)was used for quantitative detection of miRNA-192. The experimental data were statistically analyzed by the SPSS software.Results The expression levels of miRNA-192 in lung cancer tissues of patients severe smoking(over 3 cigrattes one day for more than 2 years )and drinking were significantly higher than that in patients without smoking and drinking or mild smoking(less than 3 cigrattes one day or less than 2 years). In the 47 cases, the expression of miRNA-192 had no significant difference in lung cancer tissues (△CT=8.19±2.94) and in normal lung tissues(7.42±2.64)(P>0.05). There was no correlation between the expression of miRNA-192 in lung cancer tissues and the clinical data.Conclusions MiRNA-192 is up-regulated in the population with risk factors such as smoking and drinking, and plays an role in the pathogenesis of lung cancer.There was no significant difference in miRNA-192 expression between lung cancer tissues and normal lung tissues,so we can't make sure miRNA-192 as a oncogene or suppressor gene of lung cancer. miRNA-192 can't be used in judging the pathological types,staging and prognosis of lung cancer.
