金叶女贞内生真菌分离及抑菌活性
Isolation of Endophytic Fungi from Ligustrum vicaryi and Screening of Their Antibacterial Activity

摘要: [摘要]目的 探讨金叶女贞内生真菌的多样性及其次生代谢产物抑制细菌活性.方法 采用植物组织块培养分离法从金叶女贞茎和叶中分离内生真菌,分别进行YPD液体培养,将培养物过滤后用丙酮萃取次生代谢产物,以大肠杆菌(Escherichia coli)、绿脓杆菌(Pseudomonas aeruginosa)、金黄色葡萄球菌(Staphylococcus aureus)为试验指示菌株,用滤纸片法分别测定次生代谢产物对3种供试菌的抑菌活性.结果 从金叶女贞茎和叶中共分离得到29株内生真菌,有25株内生真菌对试验菌株有一定的抑制作用,占总分离菌株的86.2%.不具抗菌活性的菌株为4株,占总分离菌株的13.8%.结论 金叶女贞茎、叶中均存在丰富的内生真菌资源,其次生代谢产物具有良好的抑菌活性.Abstract: [Abstract]Objective The study was aimed to investigate the diversity of endophytic fungi from Ligustrum vicaryi and their antibacterial activity. Methods Endophytic fungi were isolated by the method of explant culture from tissue segments and then cultured in YPD liquid medium,respectively. The fermentation broth and mycelia were extracted with acetone after filtration. Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus were selected as indicators. The extracts were tested by filtering paper method. Results The results indicated that 29 strains were isolated from L. vicaryi,and 25 of them were found to have a certain antibacterial activity(86.2% of the total strains isolated). The other 4 strains were considered inactive(17.2% of the total strains isolated). Conclusion There were abundant endophytic fungi in the stem and leaf of L. vicaryi. Their secondary metabolites showed significant antibacterial activity.
