干燥综合征蛋白组学的研究

摘要: [摘要]目的 应用SELDI研究干燥综合征疾病的特异性差异蛋白.方法 选用CM10芯片,应用表面增强激光解析离子飞行时间质谱技术,比较SS疾病组与正常组血清的蛋白及SS疾病组中关节肿痛组与无关节肿痛组血清的蛋白质表达差异.结果(1)SS组与正常组的血清中有68个差异质谱峰(P<0.05),并鉴定出11个蛋白在SS组中10个上调表达,1个下调表达;(2)未发现与关节肿痛相关的特异性差异蛋白;(3)将11个差异蛋白分别与临床资料进行分析对比,未发现明显相关性.结论(1)SS患者与正常人血清中蛋白质的表达有差异;(2)伴关节肿痛症状SS患者无特异性差异蛋白;(3)检测出的α-1抗糜蛋白酶、纤维蛋白原α链、β淀粉样蛋白前体、转甲状腺素蛋白、β2-微球蛋白都可能参与SS的发病,上述物质均可引起淀粉样变,损伤机制可能与淀粉样变相关.

Abstract: [Abstract]Objective To investigate the specific differentially expressed proteins in sjogren's syndrome by SELDI . Methods We selected the CM10 Chip and SELDI technology to compare the difference in the proteins expression between SS and health, joint swelling pain group and without joint swelling pain group. Results(1)There were 68 different mass spectrum peak between SS group and normal group(P < 0.05), and identified 11 differently expressed proteins,including 10 of upreguated and 1 downregulated protein. (2)No specific proteins were found in the joints swelling pain group. (3)The 11 different proteins were analyzed respectively with clinical data, but no correlation was found. Conclusions(1)There are difference in serum proteins express between SS patients and normal subjects.(2)The joint swelling pain symptoms in SS patients is non-specific. (3)The alpha 1 chymotrypsin,fibrinogen aa chain,beta amyloid precursor protein and thyroid hormone,β2 -microglobulin are associated with SS and can result in amyloidosis, the mechanism may be associated with amyloidosis.
