重楼地上部分与地下部分皂苷的抗肿瘤作用

摘要: [摘要]目的 探讨滇重楼地上部分和地下部分总皂苷对小鼠胃癌细胞株 (MFC)、人乳腺癌细胞株(MCF-7)和人宫颈癌细胞株(Hela)体内外生长的抑制作用.方法 采用CCK-8检测滇重楼地上部分和地下部分总皂苷对肿瘤细胞体外生长的抑制作用;将对数生长期的肿瘤细胞接种于正常小鼠左前肢腋窝皮下复制小鼠荷瘤模型.模型动物随机分为生理盐水组(NS)、环磷酰胺(30 mg/kg)组、滇重楼地上部分总皂苷高、中、低剂量(60、30、15 mg/kg)组和滇重楼地下部分总皂苷高、中、低剂量(60、30、15 mg/kg)组.分别进行腹腔注射给药,每天1次,连续15 d.观察肿瘤生长状况和小鼠生存期.结果 滇重楼地上部分总皂苷和地下部分总皂苷均可显著抑制小鼠胃癌细胞株 (MFC)、人乳腺癌细胞株(MCF-7)和人宫颈癌细胞株(Hela)的生长,并呈一定剂量和时间依赖性;滇重楼地上部分和地下部分总皂苷对模型小鼠体内肿瘤生长均有抑制作用(P<0.01或P<0.05).结论 滇重楼地上部分和地下部总皂苷均有明显的抑制小鼠胃癌细胞株 (MFC) 、人乳腺癌细胞株(MCF-7)和人宫颈癌细胞株(Hela)体内外生长的作用.

Abstract: [Abstract]Objective To investigate the anti-tumor effects of total saponins, extracted from stems and leaves of Paris polyphylla var. yunnanensis. Methods Taking mouse stomach carcinoma MFC cell line, human mammary cancer cell line(MCF-7)and human cervical carcinoma cell line(Hela),and their tumor-bearing mice as models, the antitumor activity was carried out in vitro and in vivo. CCK-8 assay was used to determine the inhibitory effect in vitro. The tumor-bearing mice model was induced by tumor cell vaccination in normal mouse forelimb left axillary subcutaneous and these mice were randomized into NS group, cytoxan group(30 mg/kg),saponins high-dose, medium-dose and low-dose groups(60,30 and 15 mg/kg). They were given intraperitoneal injection once a day for consecutive 15 days. The tumor inhibition rate and survival of the tumor-bearing mice were measured. Results Saponins, extracted from both aboveground and underground of P. polyphylla var. yunnanensis could significantly inhibit the growth of MFC,MCF-7 and Hela cells in time- and dose-dependent manners. The tumor weight in each drug treated group was significantly lower than that in the control group. Conclusion Saponins,extracted from both aboveground and underground of P. polyphylla var. yunnanensis have antitumor activity.
