甲基苯丙胺对大鼠海马DA、5-HT及TNF-α表达的影响
Effects of Methamphetamine on the Expression of DA, 5-HT and TNF-α in Rats Hippocampus

摘要: 目的 研究甲基苯丙胺 (methamphetamine, MA) 对大鼠海马多巴胺 (Dopamine、DA) 、5-羟色胺 (5-Hydroxytryptophan, 5-HT) 及肿瘤坏死因子-α (tumor necrosis factor-α, TNF-α) 的影响.方法 (1) 通过连续腹腔注射甲基苯丙胺15 mg/ (kg·次) , 5 d, 每天2次, 建立甲基苯丙胺体内中毒模型.采用甲基苯丙胺伴药培养混合海马神经元/小胶质细胞24h, 建立甲基苯丙胺体外中毒模型; (2) 采用酶联免疫法检测大鼠海马及细胞培养上清液多巴胺 (DA) 、5-羟色胺 (5-HT) 及肿瘤坏死因子α (TNF-α) 表达的变化.结果 在第5天, 甲基苯丙胺处理大鼠毛发树立, 体重下降, 快速的摇头、转圈或头的背腹运动, 刻板行为评分5分.其海马DA (P<0.05) 、5-HT (P<0.05) 水平下降及TNF-α含量升高 (P<0.05) .甲基苯丙胺处理混合海马神经元/小胶质细胞24 h细胞培养上清液DA (P<0.05) 、5-HT (P<0.05) 水平下降及TNF-α含量升高 (P<0.05) .结论DA、5-HT及TNF-α的变化参与了甲基苯丙胺对大鼠海马的损伤.Abstract: Objective To study the effect of methamphetamine on the expression of DA, 5-HTand TNF-αin rats hippocampus. Methods (1) The in vivo toxicity model was established by injecting MA 15 mg/ (kg.time) , two times a day for 5 days. The in vitro toxicity model was established by incubating MA with mixed hippocampal neurons/microglia for 24 hours. (2) ELISA was used to detect the content of DA, 5-HT and TNF-α in rat hippocampus and cell culture supernatant. Results On the fifth day, Methamphetamine treated rats showed hair errecting, weight loss, rapid head shaking, circling or head dorsal movement, with a stereotyped score of 5. The levels of DA and 5-HT decreased, and the content of TNF-alpha elevated in rats hippocampus of MA group. The levels of DA, 5-HT decreased and the content of TNF-alpha increased in cell cultural supernatants of MA group24 hours. Conclusion The changes of DA, 5-HT and TNF-alpha are involved in rat hippocampal damage induced by methamphetamine.
