Abstract:
[Abstract]Objective To explo the antioxidant effect and molecular mechanism of gastrodin(Gas) in epilepsy(EP) rats induced by LiCl-pilocarpine(PILO). Methods Eighty male SD rats were randomly divided into 5 groups: sham group, EP group, therapy groups(pretreated with 60 mg/kg, 90 mg/kg, 120 mg/kg of gastrodin respectively). The EP model was esteblished by peritoneal injection of LiCl-PILO. Therapy groups were pretreated with various concerntration of Gas. The control group was given the same dosage of normal saline. The alteration of behavior was observed, the concentration of catalase(CAT),glutathion(GSH), superoxide dismutase(SOD),glutathion reductase(GR), total antioxidtion(T-AOC) and malondialdehyde(MDA) in rats brain cortex were detected by chemical colorimetric method, phosphorylation of p38 was determined by western blot. Results There was no EP seizure in sham group, and the EP seizure degree in therapy groups(gas pretreated groups) was significantly decreased, and had statistically significant difference with EP group(P<0.05). The EP model rats exhibited a significant decrease in the concentration of endogenous antioxidants(CAT, GSH, SOD, GR and T-AOC),while an increase of the concentration of MDA and phosphorylation p38 protein as compared to sham group(P<0.05). After treatment of the Gas,treatment group rats attenuated the seizure degree, exhibited a significant increase of the concentration of endogenous antioxidants(P<0.05),while a decrease in concentration of MDA and phosphorylation of p38 as compared to model group(P<0.05). Conclusion Gas may have a neuroprotective role in central nervous system of epileptic rats modle by down-regulateing the seizure degree and the activity of p38 kinase and up-regulateing the content of endogenous antioxidants.

Abstract:
[Abstract]Objective To investigate the effect of geraniin on expression of Wnt3a protein and mRNA in bone marrow stromal cell (BMSC) from osteoporotic rats. Methods The model of osteoporosis (OP) was duplicated by ovariectomy in rats. BMSCs were isolated and cultured. BMSCs from shamed rats were routinly cultured and taken as normal control, and BMSCs from OP rats were divided into model group, 1 μmol/L simvastatin positive group, and geraniin group (0.01, 0.1, 1, 10, 100 μmol/L), respectively. The methods of realtime-PCR and western blot were used to assay the protein and mRNA expression of wnt3a, respectively. Results As compared with normal control group, the protein and mRNA expression of wnt3a in model group were significantly suppressed; Compared with model group, 1 μmol/L simvastatin, and 0.1, 1, 10 and 100 μmol/L geraniin significantly increased the expression of wnt3a protein and mRNA. Conclusion It is suggested that geraniin activates wnt/β-catenin pathway though increasing the expression of signaling protein wnt 3a in BMSCs from OP rats. It would be beneficial to osteogenic differentiation of BMSCs and osteogenesis.

Abstract:
[Abstract]Objective To investigate the molecular mechanism of Loureirin A mediated anti-hepatic fibrosis by evaluting its effects on proliferation ,secretion of α-smooth muscle actin(α-SMA)and transforming growth factor-beta1(TGF-β1),and expression of rat hepatic stellate cells in vitro . Methods Primary hepatic stellate cells were isolated and cultured from Sprague-Dawley rats. After activating and inducing primary hepatic stellate cells from qHSC to aHSC,the activated hepatic stellate cells model in vitro was established. Then we observed the morphological changes of static hepatic stellate cells and activated hepatic stellate cells with inverted phase contrast microscope. Cultured hepatic stellate cells were treated with different concentrations of loureirin A and the inhibitory rate of HSCs proliferation was measured by MTT assay. The expression of Frizzled-4 was measured by western blot analysis. The content of α-SMA and TGF-β1 in the cultured HSCs'supernatant were measured by enzyme-linked immunosorbent assay(ELISA) . Results Loureirin A the proliferation of inhibited activated hepatic stellate cells in a time-dose-dependent manner compared with the control group,IC50=0.30 μg/μL. After loureirinA treatment of the HSCs,western blot analysis showed that Frizzled-4 expression level was obviously lower than control group. Loureirin A also inhibited α-SMA and TGFβ1(P<0.05)secretion in the cultured HSCs'supernatant in different degree by the assay of ELISA. Conclusions The molecular mechanism of Loureirin A and Wnt signaling pathway mediated anti-hepatic fibrosis and anti-angiogenesis may involve down-regulation the expression of Frizzled-4,inhibiting the synthesis and secretion of α-SMA,TGF-β1and the proliferation of HSCs.
