Expression of Astrocyte, Neuron, and Proinflammatory Cytokines Derived from Frontal Cortex of Methamphetamine Dependent rats after Gastrodin Administration

Abstract:
Objective To observe the curative mechanism and effect of neurotoxicity injury induced by methamphetamine (MA) and the neuroprotective effects of gastrodin interfered. Whether the expression of astrocyte and proinflammatory cytokines has contributed to the effects of gastrodin.Me thods 48 healthy male SD rats were randomly divided into three groups: control group (Daily intraperitoneal injection of saline for 8 weeks) , MA group (A dose of 10 mg/kg MA was administered every day for four weeks, then given daily intraperitoneal injection with10 mg/kg saline for 4 weeks) and gastrodin group (A dose of 10 mg/kg MA was administered every day for four weeks, then given daily intraperitoneal injection with 10 mg/kg gastrodin for 4 weeks) . The behavioral changes of rats were measured by conditioned place preference ( CPP) and sterotyped behavior ( SB) induced by methamphetamine. Immunofluorescence staining was used to detect the expression of glial fibrillary acidic protein (GFAP) and NEUN in rat frontal cortex.The expression of IL-6 and TNF-α were detected by quantity RT-PCR and westrn bloting.Re s ults Compa MA depndent 4 weeks group with control group, the scores of sterotyped behavior of MA depndent groups had signficantly increased (P <0.01) . Comparing MA depndent 4 weeks group with MA depndent 4 weeks+gastrodin group, the scores of sterotyped behavior of MA dependent 4 weeks group had obviously decreaseed (P<0.01) . Compared with the control group, the expression of GFAP of MA dependent 4 weeks group decreased and the expression of NEUN increased. Compared MA dependent 4 weeks group with control group, the expression of IL-6 and TNF-α increased (P < 0.01) . Compared MA dependent 4 weeks+gastrodian group with MA dependent 4 weeks group, the expression of TNF-α and IL-6 significantly reduced (P < 0.01) .Conclus ion The neurological damage induced by methamphetamine might be related to the activation of astrocytes and the high expression of inflammatory cytokines including IL-6 and TNF-α. Gastrodin could abate the neurological injury of methamphetamine dependence via reducing the activation of astrocytes and decreasing the expression of IL-6 and TNF-α.

Abstract:
Objective By comparing with the SD rats mixed glial cells, C57 mice mixed glial cells and Kunming mice mixed glial cells and exploring the expression of inflammatory factor of three mixed glial cells induced by lipopolysaccharide (LPS) , the study aimed to explore the application value of three kinds of mice and find the ideal model of inflammation. Me thods We used LPS as inducers, and NO, IL-1β, COX-2 and i NOS as anti-inflammatory antioxidant index. After cutting the head and taking the brain, we cultured the mixed glials. Then we used Greiss assay to detect the expression of NO and used western blot to detect the expression of protein of IL-1β, COX-2, and i NOS protein.Finally we compared the mixed glial cells from SD rats, C57 mice mixed glial cells and Kunming mouse mixed glial cells and selected the best inflammation model from three mixed glial cells.Re s ults The results showed that the morphological changes of the mixed glial cells in SD rats were treated with N2-free medium. Compared with the control group, the quantity of NO of LPS group of three mixed glial cells increased significantly (P<0.01) . The LPS group of SD rats released the highest concentration of NO. Western blot was used to detect the expression of IL-1β, Cox-2 and i NOS in three kinds of rodents. Compared with the blank control group, the expression of COX-2 protein, i NOS and IL-1β in the LPS group of the three mice increased significantly. The results showed that LPS could successfully stimulate the release of inflammatory cytokines in three kinds of mice, among which the SD rats were more sensitive and it could be used in the study of AD inflammation model. Conclus ion The results showed that LPS could induce the release of NO and the expression of IL-1β, i NOS and COX-2 in C57 BL/6 mice, Kunming mice and SD rats to induce inflammatory response. Thus, LPS can induce the formation of inflammatory oxidation models of the original mixed glial cells of the three mice. Moreover, the SD rats were more sensitive.

Abstract:
Objective To explore the protective effect of gallic acid in Phyllanthus emblica on high glucose-induced apoptosis of pancreatic islet β cells, and to provide a reference for the discovery of natural compounds for the treatment of diabetes. Me thods In vivo experimental model, wistar male rats were used as in vivo subjects and 50 mg/kg STZ was injected intraperitoneally. After the model was successfully established, 25 mg/kg of gallic acid was given orally, and the positive drug was Sitagliptin. After 4 weeks of administration, theblood was taken and the pancreas was removed for HE staining. Western blot was used to measure the expression of NLRP3 and TXNIP in pancreatic tissue in high sugar state. In vitro model, insulinoma cell line INS-1 cells were used as in vitro targets to establish high levels. In sugar-induced apoptosis model, INS-1 cells were cultured in glucose-free RPMI 1640 complete medium supplemented with 25 mmol/L glucose. Gallic acid was used as the test sample. Experiments were divided into normal controls, high-sugar models, and low, medium and high levels of gallic acid groups. The cell viability was measured by MTT assay. The m RNA expression of NLRP3 and TXNIP in INS-1 cells was detected by QPCR and Western blot, and the expression of NLRP3 and TXNIP protein was detected.Re s ults (1) INS-1 cells were cultured in a medium with glucose concentration of 25 mmol/L for 48 h, and the apoptosis rate was increased compared with the control group (P <0.01) , indicating that the apoptosis model was established successfully under high glucose conditions. (2) 10, 5, and 2.5 μmol/L GA were used to treat the control group and the high glucose model group cells respectively. The survival rate of the control group did not change significantly (P>0.05) . Compared with the control group, the expression of NLRP3 and TXNIP in INS-1 cells in the high glucose model group was significantly different (P<0.05) ;the protein expression level was significantly downregulated after GA treatment, and there was a statistical difference (P<0.05) . Compared with the control group, the expression of NLRP3 protein in INS-1 cells in the high glucose model group was statistically different (P<0.01) , and the protein expression level was significantly downregulated after GA treatment (P<0.01) ; The protein expression level was up-regulated (P<0.05) ;the protein expression level after GA treatment was significantly down-regulated (P<0.05) ; (4) The expression of NLRP3 and TXNIP m RNA in INS-1 cells was increased in the high glucose model group compared with the control group (P <0.01) ; The expression of protein was significantly down-regulated after GA treatment (P<0.01) . Conclus ion The cells were cultured for48 h in glucose-free RPMI 1640 complete medium supplemented with 25 mmol/L glucose. GA has no effect on the proliferation of normal INS-1 cells. GA protects INS-1 cells from apoptosis under high glucose conditions. The mechanism may be related to GA down-regulation of NLRP3 and TXNIP gene expression.
“The Window Effect” of 1800 MHz Electromagnetic Radiation on Oxidative Stress in Skin of SD Rats

Abstract:
Objective To investigate the effect of 1 800 MHz electromagnetic radiation on activity of SOD and GSH-Px in the skin tissues of SD rats.Me thods A total of 98 healthy SD rats with SPF level, aged 4 weeks, were randomly divided into radiation group and control group.The radiation group was totally exposed under 1 800 MHz electromagnetic wave with seven different power density of radiation of 0.1 m W/cm2, 0.3 m W/cm2, 0.5 m W/cm2, 0.7 m W/cm2, 0.9 m W/cm2, 1.0 m W/cm2 and 1.2 m W/cm2 respectively.It lasted 21 days and for a period of 12 hours a day. After radiation, the activity of SOD and GSH-Px in the skin tissues were detected by enzyme marker.Re s ults In radiation group, the activity of SOD and GSH-Px in the skin tissues of SD rats were decreased under0.3 m W/cm2 and 0.5 m W/cm21 800 MHz electromagnetic wave. Compared with the control group, there was a significantly difference in radiation group (P<0.05) .While under other four 1 800 MHz electromagnetic waves, the activity of GSH-Px and SOD in the skin tissues showed no statistical difference between the two groups (P>0.05) .Under 1 m W/cm21 800 MHz electromagnetic wave, the activity of GSH-Px showed no statistical difference between two groups ( P >0.05) . Conclus ion The power density of 0.3 m W/cm2 and 0.5 m W/cm2 1 800 MHzelectromagnetic wave can reduce the activity of GSH-Px and SOD in the skin tissues of rats.

Abstract:
Objective To study the effect of Astragalus polysaccharide on the expressions of Nephrin and Desmin in high sugar stimulation mice podocytes. Me thods Test one: the cultured mouse podocytes were divided into 5 groups: astragalus polysaccharide group [D-glucose 30 mmol/L + astragalus polysaccharide (0.0 g/L, 0.1 g/L, 0.2 g/L, 0.4 g/L and 0.8 g/L) ]. Test two: In test one, we chose the concentration of astragalus polysaccharide which most affected both expression of Nephrin and Desmin in podocytes and named it as astragalus polysaccharide 0.X g/L. The cultured mouse podocytes were divided into 7 groups: astragalus polysaccharide group (D-glucose 30 mmol/L + astragalus polysaccharide 0.X g/L) had been cultured for 0 h, 3 h, 6 h, 12 h, 24 h, 48 h and 72 h, respectively. Test three: Basted on test two, we chose the time which most affected both expression of Nephrinand Desmin in podocytes and named it as Yh. The cultured mouse podocytes were divided into 4 groups: the control group, mannitol hypertonic glucose control group, high sugar group, and astragalus polysaccharide group (high sugar group + astragalus polysaccharide 0.X g/L) . Each group had been cultured for Yh. Nephrin and Desmin of each subgroup was detected by flow cytometry podocytes and real-time PCR. Re s ults With the intervention of polysaccharide, Nephrin in podocyte cell surface gradually increased, Desmin gradually reduced in a concentration-dependent manner ( P <0.05) . With the intervention of astragalus polysaccharide, Nephrin gradually increase and Desmin expression gradually decreased in a concentration-dependent manner (P<0.05) .Compared to the control group and the high sugar group, Nephrin mR NA increased and Desmin mR NA decreased in0.8 g/L astragalus polysaccharide co-cultured for 72 h ( P <0.05) . Conclus ion Astragalus polysaccharide intervention can increase Nephrin and reduce Desmin in podocytes, which are possible targets of Astragalus polysaccharide in treatment of DN proteinuria.

Objective Veratraldehyde is an important pharmaceutical intermediate and chemical raw material. Taking common synthetic methods of Veratraldehyde into consideration, the study is aimed to further investigate a more efficient synthetic route in order to provide a solid foundation for the application of Veratraldehyde.Me thods This article introduces an alternative method based on the Triton-B catalyzed olefin isomerization of methyl Eugenol followed by ozonation/reduction reaction.Re s ults In the experiment, the optimal conditions was selected by investigating the influence of reaction time, reaction temperature and catalyst dosage on the yield of Veratraldehyde. The total yield of this 2-step reactions was 64%. Conclus ion The above-mentioned synthesis method only requires raw materials that are easily acquired and its relative reaction conditions are mild, thus it will provide values in the industrial applications.

Abstract:
Objective To study antioxidant role and mechanism of Rg1 in rats with cerebral ischemia reperfusion injury (IRI) .Me thods One hundred and twenty healthy male rats were randomly divided into six groups: control group, sham operation group, model group, different concentration (30, 60, 90 mg/kg) of Rg1 treatment group.MCAO SD rats model was established by suture-occluded method;the Rg1 treatment groups were given Rg1 i.p. in advance, after 24 hours of reperfusion, neurobehavioral scores of all groups were examined by Longa's standard;The expression of Nrf2 and HO-1 were analyzed by western blot;The content of SOD and MDA was detected by kit.Re s ults The score of model group rats are significantly higher than control group, compared with the model group, the score of different concentration of Rg1 treatment group was decreased (P<0.05) . The Nrf2 and HO-1 expression in model group was mildly higher than the control or sham group (P <0.05) . Both of them in every Rg1 treatment group was higher than model group. Compared with control or sham group, SOD content was observably depressed but MDA content was dramatically increased in model group, interestingly, SOD contentwas enhanced, MDA content was attenuated in different concentration of Rg1 treatment group (P <0.05) .Conclusion Rg1 increases Nrf2 and HO-1 protein expression and SOD content, reduces MDA content, improves neurofunctional performance of rats after MCAO, and alleviates cerebral cerebral IRI.
Pyrethroid Exposure and Related Factors among Pregnant Women in the First Trimester in Rural Yunnan Province, China

Abstract:
Objective To investigate the level and relevant factors of pyrethroid pesticide exposure among pregnant women during the first trimester in rural areas of Yunnan province, and to provide basic data for further study and provide a basis for the formulation of intervention measures to reduce exposure.Me thods According to the birth cohort of rural Yunnan built in 2014, pregnant women in the first trimester were recruited in two townships of an agricultural county who were confirmed pregnant and continued pregnancy in antenatal clinics. The concentration of the metabolites of the pesticides in the urine of women was determined by super high performance liquid chromatography and the contact status of pyrethroid pesticides was analyzed using a questionnaire survey.Re s ults A total of 419 women in the first trimester were surveyed and 94.51% of women detected at least one of the pyrethroid metabolites, among which the detection rates of 3-Phenoxybenzoic acid (3-PBA) , cis-/trans-3- (2, 2-Dichloroethenyl) -2, 2-dimethyl-cyclopropanecarboxylate (cis-/trans-DCCA) and 4-fluoro-3-phenoxybenzoic acid (4-F-3-PBA) were 79.47%, 62.05% and 24.58%, and the median concentrations were3.53 ng/m L, 14.80 ng/m L and less than the minimum detection limit 0.4 ng/m L.Only 12 people (2.95%) reported that they had exposure to pesticides.Self-reported exposure rate was not consistent with the actual exposure rate.Multivariate linear regression analysis showed that the season was a factor affecting the level of three metabolites in women in the first trimester. Higher lever was found in women in spring (t=2.147, P=0.032) , summer (t=2.144, P=0.035) and winter (t=2.453, P=0.015) compared to those in autumn.Conclus ion Pyrethroid pesticides were widely exposed in the pregnant women in rural areas of Yunnan province. Some women have higher exposure level, which is related to the season.
The Expression of LRG1 in Cervical Squamous Carcinoma and Its Relationship with Microvessel Density

Objective To investigate the expression of LRG1 (Leucine-rich alpha-2-glycoprotein 1) and CD105 (Endoglin) in normal cervical tissues, cervical intraepithelial neoplasia (CIN) Ⅱ-Ⅲ and cervical carcinoma. Furthermore, to explore the function of LRG1 in cervical carcinoma pathogenesis and the relationship between LRG1 and microvessel density.Me thods The expression levels of LRG1 and CD105 were detected by immunohistochemistry in 40 cervical carcinoma tissues, 20 CIN Ⅱ-Ⅲ tissues and 20 normal cervical tissues.Re s ults Compared to the normal cervix, the expression of LRG1 in CIN Ⅱ-Ⅲ and cervical squamous cell carcinoma was significantly increased (P<0.05) .LRG1 expression was correlated to clinical stage and lymph node metastasis (P <0.05) . But there was no correlation between LRG1 expression and age, the size of tumor, pathologic grades and depth of invasion (P > 0.05) .With the deepening of cervical lesions, CD105-MVD (X±s) increasedsignificantly (P<0.05) .The expression of CD105-MVD in cervical carcinomawas related to clinical stage, lymph node metastasis and the size of tumor. The difference was statistically significant (P<0.05) . There was no correlation between CD105-MVD and age, pathologic grades and depth of invasion (P > 0.05) .Positive correlation was found between the expression of LRG1 and CD105-MVD (r = 0.944, P <0.05) .Conclus ions The expression of LRG1 and CD105-MVD is up-regulated and shows a positive correlation, which suggests that the abnormal expression of LRG1 and CD105-MVD may involve in the occurrence and development of cervical squamous carcinoma.
Effect of Collagen Thread in Tension-Free Suture of Oral Implant Surgical Incision and Effect on Wound Healing Time

Abstract:
Objective To investigate the effect of collagen line in the tension-free suture of the incision of oral implant and the effect on the healing time of the wound. Me thods 100 patients were selected from January2016 to January 2017 for repair of dental implant in Zhong Hua Dental clinic of Jinghong city, and they were randomly divided into control group and study group, with 50 cases in each group. The control group was treated with non absorbable sutures of silk suture wounds, while the study group using collagen suture wounds, then we compared two groups of patients with wound healing grade, suture line absorption, wound healing time and formation rate. Re s ults The research group of grade A healing rate was 96%, significantly higher than 80% in the control group (P < 0.05) , and the adverse reactions of the study group was 6%, lower than 20% in the control group (P < 0.05) ;the complete absorption rate and smooth incision rates in the study group for suture in different times were 84%, 96% 100%, and 86%, which were significantly higher than the control group's 62%, 78%, 80% and 68% (P < 0.05) . The wound's healing time and postoperative VAS score of 3 d, 5 d and 7 d were significantly lower than the control group (P < 0.05) . Conclus ion Collagen line in oral implant surgery incision tension-free suture of wound healing and postoperative recovery is significantly better than that of silk and non absorbable sutures, and collagen can be degradated orally, which effectively improves the wound healing of patients with grades and wound flat rate, and effectively accelerates wound healing, with the value of application.
The Changes of Hepatitis C Virus Gene Subtypes and RNA Loads in the Development of Chronic Hepatitis C Patients in Yunnan

Abstract:
Objective To investigate the changes of hepatitis C virus (HCV) gene subtypes and RNA loads in chronic hepatitis C, HCV-related liver cirrhosis and HCV-related hepatocellular carcinoma patients in Yunnan. Me thods 241 patients were enrolled at the First Affiliated Hospital of Kunming Medical University (Yunnan, China) from January 2016 to April 2017. Among them, 169 patients were with chronic hepatitis C, 56 patients with HCV-related liver cirrhosis and 16 patients with HCV-related hepatocellular carcinoma. HCV gene subtype and RNA loads were measured using the Polymerase Chain Reaction-fluorescence probe method. Re s ults In the chronic hepatitis C group, there were 47 subtype 3 b cases (27.81%) . 17 cases of HCV-related liver cirrhosis were subtype 1 b (30.36%) ;5 patients with HCV-related hepatocellular carcinoma were subtype 3 b (31.25%) .There was no statistical difference distribution of the genotype among the three groups (P>0.05) .The HCV RNA loads of the chronic hepatitis C group, HCV-related liver cirrhosis group and HCV-related hepatocellular carcinoma group were (332±114) copies/m L, (189±73) copies/m L and (152±56) copies/m L respectively.The difference among three groups were significant (P <0.01) .The chronic hepatitis C group was significantly higher than the other two groups, and the differences were statistically significant (P<0.01) . But no significant difference of HCV RNA loads was found between HCV-related liver cirrhosis and HCV-related hepatocellular carcinoma group (t = 0. 65, P<0.05) . Conclus ion In Yunnan, 3 b was main genotype in chronic hepatitis C patients and 1 b was main genotype in HCV-related liver cirrhosis, 3 b was main genotype in HCV-related hepatocellular carcinoma. HCV RNA loads tend to decrease in the progress that chronic hepatitis C develops into HCV-related liver cirrhosis and HCV-related hepatocellular carcinoma.

Abstract:
Objective To explore the clinical efficacy of Montelukast in the treatment of severe asthma.Me thods A retrospective analysis was conducted in the Department of Respiratory Medicine from January 2016 to August 2017.A total of 168 patients with moderately severe asthma were randomly divided into control group and observation group. The control group was treated with leukotriene receptor antagonist Montelukast and the observation group was treated with both Montelukast and budesonide inhalation suspension. Then we compared the clinical effect, lung function and blood gas index before and after treatment between the two groups. Re s ults Significant improvement was observed in the clinical symptoms and lung function in the two groups of patients after the treatment.Compared to the control group, a better improvement was seen in the observation group (P < 0.05) . The results of blood gas analysis were significantly different between the two groups before and after the treatment (P < 0.05) .Conclus ion In the treatment of moderately severe asthma, Montelukast combined with budesonide inhalation suspension has good clinical effect and can significantly alleviate symptoms, which can be widely used clinically.

Abstract:
Objective To study the neuro electrophysiological characteristics of paraneoplastic peripheral neuropathy (PPN) . Me thods A retrospective study was conducted for 29 PPN patients consecutively referred to Neurology Department of the First Affiliated Hospital of Kunming Medical University between January 2000 and June2017. The electrophysiological characteristics of motor nerves, sensory nerves of upper and lower limbs were analyzed. Measurement indicators include: (1) The motor conduction velocity and compound muscle action potential amplitude of median, ulnar, tibial, and common peroneal nerves; (2) The sensory conduction velocity and sensory nerve action potential amplitude of median, ulnar, tibial, and superficial peroneal nerves; (3) F waves of median and tibial nerves.Results (1) For patients with PPN, their motor and sensory nerves in upper and lower limbs were damaged. The total anomaly rate of the amplitude was higher than that of the nerve conduction velocity (P<0.05) , while the abnormal rate of amplitude of sensory nerve action potential was greater than that of motor nerve compound muscle action potential (P<0.05) . Abnormal motor nerve conduction velocityhad a similar incidence to abnormal sensory nerve conduction velocity (P>0.05) . (2) Nerve conduction study showed that abnormality rate of lower extremity is higher than that of upper extremity. (3) Abnormal F waves were observed less frequently than abnormal nerve conduction rates (P<0.05) . Conclusions The electrophysiological properties of PPN were frequently seen in sensorimotor neuropathy. The damage of distal extremities is more serious.The damage in lower extremity is more severe than that in the upper extremity. The axonal damage mainly occurred in sensory nerves. There is no obvious difference in the degree of demyelination between motor and sensory nerves.Evidence can be provided by analyzing the neuro ectrophysiological characteristics for diagnosis of paraneoplastic peripheral neuropathy in early stage.

Abstract:
Objective To improve the prognosis among patients with respiratory failure poisoned by organophosphorus pesticide though analyzing and discussing the emergency measures and treatment effects. Me thods From January 2014 to December 2016, 86 patients with respiratory failure caused by severe organophosphorus pesticide poisoning were received in the Emergency Department in Nanchong Central Hospital.The patients were numbered according to the order of the treatment and randomly divided into control group and observation group with43 cases in each group.All patients were treated with routine emergency measures (gastric lavage and endotracheal intubation, rehydration, and symptomatic treatment) .The control group was treated with pralidoxime chloride injection and injection of atropine detoxification while the observation group was given pralidoxime chloride combined with Penehyclidine Hydrochloride Injection detoxification.We compared the alleviation of clinical symptoms and the changes of cholinesterase (CHE) and respiratory function (respiratory frequency, Pa02, Pa02/Fi02) between the two groups of patients 10 min and 30 min after administration. The statistics of atropinization time, blood purification treatment rate, tracheotomy rate, hospital mortality, complications and treatment time were recorded.Re s ults No statistic significance was observed among the two groups of patients in gender, age, weight, body mass index (BMI) , the severity of poisoning, types of organophosphorus drugs and blood cholinesterase (CHE) at the first visit (P > 0.05) .Blood CHE was effectively improved among the two groups 10 min and 30 min after the treatment and significantly higher CHE was seen in the observation group compared to the control group (P <0.05) . The overall clinical symptom rate was lower 10 min and 30 min after the treatment (P < 0.05) , and the clinical symptom rate of the observation group was lower than that of the control group (P < 0.05) . The respiratory function indexes (respiratory rate, Pa02, Pa02/Fi02) were significantly improved 10 min and 30 min after the treatment in both groups compared with those before the treatment (P < 0.05) .The blood purification treatment rate, tracheotomy rate, complication rate, hospitalization time and atropine time of the observation group were significantly lower than those of the control group (P < 0.05) . No significant difference was found in mortality rate between the two groups (P >0.05) .Conclus ion After giving effective respiration and circulation support, administration of penehyclidine hydrochloride combined with pralidoxime chloride detoxification can effectively restore the cholinesterase activity among patients with organophosphorus pesticide poisoning and improve respiratory function and prognosis, whichshows a high clinical value.

(3) The value difference of Number Cancellation Test One between pre and post treatment of risperidone was related with different genotypes of two polymorphismsin NRG1 gene, there was statistically difference in two genotypes of rs35753505 loci: G/A group was higher than G/G group (P = 0.010) and in three genotypes of rs3924999 loci: A/A group was higher than A/G group (P = 0.032) . (4) The value difference of Reven's Standard Progressive Matrices between pre and post treatment of risperidone was related with rs35753505 polymorphic loci: G/A group was higher than G/G group (P = 0.004) (5) The result of correlation regression analysis between pre and post treatment of risperidone showed that the value difference of Number Cancellation Test Two was related with the baseline clinical data of course, dose, immediate memory, net score of Number Cancellation Test Two, Number Cancellation Test Three and the genotypes of rs35753505 loci.Conclus ion After 12 weeks treatment of risperidone: the improvement degree of patients' attention directivity and concentrated force in different genotypes of rs35753505 loci was different:G/A group was higher than G/G group.

Objective To provide evidence for the relationship between TMD and occlusal trauma by studying the mechanics influence and displacement of condyle by building the three dimensional finite element analysis model andstimulating the abnormal bite force leading occlusal trauma.Me thods We collected DICOM date of healthy male volunteers with normal occlusal relationship using CBCT scan with constraint on condylar top and formed the finite model with divide mesh.Two conditions were built: (1) We operated different vertical load on the left mandibular molar occlusal surface; (2) We operated different buccal direction load on the same modelto study the mechanic change on left condylar.Re s ults When the three dimensional finite model of mandible was built, the mechanic region of condyle stayed the same but the stress and tension increased accordingly as the loda duplicate rose. As the same buccal operation load was operated, the stress and tension became larger and the displacement became longer.Conclus ion The abnormal bite force causing occlusal trauma can change mechanic conduct on condyle and lead to the pathological change of condyle.

Objective To investigate the treatment evaluating value of ESAT-6 and CFP-10 in T-SPOT.TB kit for tuberculous vertebral osteomyelitis.Me dthods This retrospective study analyzed 29 cases diagnosed as TVO in the First Affiliated Hospital of Kunming Medical University from January 2013 to January 2016. All cases were the first-time consultancy. The Wilcoxon-singed-rank-test and chi-square test were used to analyze the changes of ESAT-6 and CFP-10 in the procedure of treatment. The linear-regression analysis was used to analyze the relationship between ESR, CRP, VAS and two specific antigens.Re s ults The pretherapeutic spot counts of ESAT-6 and CFP-10 were compared with the first and the last follow-up respectively. The results of two antigenic spots change showed a higher consistency (P<0.05) .The positive rates of CFP-10 at the prior treatment, the first follow-up and the last follow-up were 86.20%, 79.31% and 58.62% respectively. The result of chi-square test showed a higher consistency (P <0.05) . ESAT-6 only correlated with VAS. CFP-10 had the relationship with VAS and ESR. But all of these relativities were weak.Conclus ion The decrease in the spot counts of ESAT-6 and CFP-10 suggest that the treatment is effective, and CFP-10 may be one available index to evaluate the treatment effect.

Objective To investigate the relationship between the location of thyroid papillary microcarcinoma and cervical lymph node metastasis. Me thods The clinical data of 334 cases of thyroid papillary microcarcinoma were retrospectively analyzed. The location of the lesion, the size of the lesion, the situation of the capsule and other factors were compared in patients with central lymph nodes or/and cervical lymph node metastasis.Re s ults The lymph node metastasis rate were 33.33% (26/104) , 39.81% (43/108) , and 52.46% (64/122) , when the papillary thyroid microcarcinoma was located at the upper pole, median pole, and lower pole respectively (P<0.005) . There were 46 cases (46/334, 13.77%) with cervical lymph node metastasis. The metastasis rate of cervical lymph node were 21.15% (22/104) , 11.11% (12/108) , and 9.83% (12/122) , when the papillary thyroid microcarcinoma was located at the upper pole, median pole, and lower pole respectively (P = 0.003) . 3 patients (6.5%) had no central lymph node metastasis and direct skipping of the cervical lymph nod trensfer. The rate of tumor break through through the membrane in patients with cervical lymph node metastases was 30.43% (14/46) , which was higher than the 19.09% (55/288) of patients without lymph node metastasis (P <0.005) .Conclus ions There is a certain correlation between the location of papillary thyroid microcarcinoma and cervical lymph node metastasis.

Objective To investigate the risk factors of renal inadequacy after postpartum hemorrhage.Me thods According to the diagnostic criteria of postpartum hemorrhage, 200 cases of postpartum hemorrhage who were admitted to the Intensive Care Unit from January 2010 to December 2014 were collected. The general situation, history of pregnancy induced hypertension and gestational diabetes mellitus, ASA classification, anesthesia method, delivery mode, postpartum hemorrhage and other indicators. The risk factors were analyzed by logistic regression.Re s ults Among 200 cases of postpartum hemorrhage who were admitted in the intensive care unit, 21 cases with renal insufficiency were seenafter delivery (10.5%) aged between 20 and 44 years old with the average age of (29.24 ±4.58) years old. Using the ASA evaluation criteria to evaluate the preoperative situation, we observed179 cases in ASA grade level I (89.5%) , 20 casesin level II (10%) , one casein level III (0.5%) and no case in grade IV. Among all the women, we found 147 cases of vaginal delivery (73.5%) , 53 cases of cesarean section (26.5%) , 196 cases of spinal anesthesia (including labor analgesia) (98%) and 4 casesof general anesthesia (2%) . There were 12 patients with hypertension during pregnancy (6%) , and 188 patients without hypertension (94%) . There were 9 cases of gestational diabetes mellitus (4.5%) , and 191 patients without gestational diabetes mellitus (95.5%) .Conclus ions The risk factors of renal insufficiency after postpartum hemorrhage were: age older than 35 years, cesarean delivery, pregnancy history of hypertension or gestational diabetes mellitus.

Objective By comparing the methylation status of imprinted gene H19 in three different quality spermatozoa and the methylation status of the embryos after the combination of spermatozoa, the sperm methylation status in the ART technique was preliminarily explored with the postnatal embryo methylation.Me thods In this study, a total of 91 spermatozoa and 91 low-quality embryos were collected from IVF-ET in the First People's Hospital of Yunnan Province. Among them, sperm samples were divided into three groups according to the sperm concentration parameter in the WHO fourth edition, then the relationship between different quality spermatozoa and postnatal embryo methylation were analyzed.Re s ults The abnormality rate of H19 methylation status in group B was significantly higher than that of the sperm group and the embryo group.Conclus ions The abnormalities of H19 methylation mainly concentrated in the abnormal parameters of spermatozoa, suggesting that the abnormal state was related to the decrease of sperm quality;Through IVF, ICSI fertilized embryos have the same proportion of the corresponding abnormal state with no significant difference; Abnormal sperm embryos did not show abnormality, which may be related to the self-repair function of the embryo itself.

Abstract:
Objective The study discusses the clinical efficacy, surgical technique and new technology application of “ ⊥ ” incision in tracheotomy. Me thods From March 2014 to December 2015, a total of 300 patients treated in the First Affiliated Hospital of Kunming Medical University with tracheotomy prospective were studied.The statistics of success rate of incision, balloon cuff rupture and complications of “ ⊥ ” form incision (group A) and“I”form incision (group B) during tracheotomy.Result In group A, 144 out of 150 cases (96%) were inserted successfully for the first try, while the first incision was successful in 90/150 cases of group B (60%) (P<0.01) . Balloon cuff rupture was found in 3/150 cases (2%) in group A and 15/150 cases (10%) in group B (P <0.05) . The postoperative complications (postoperative bleeding, subcutaneous emphysema, pneumothorax, laryngotracheal stenosis) were observed in 10/150 cases (6.7%) in group A and 25/150 cases (16.7%) in group B (P<0.05) .Conclusion The“⊥” tracheal incision is simple, safe and causes fewer complications. It is worthy of generalization and application.

Objective To observe the normal anatomy and variation of cystic duct by MRCP, and to explore the correlation between anatomic variation and gallbladder stones. Me thods To have a retrospective analysis of 371 MRCP patients which meet the requirement of imaging, and to compare the differences between anatomic variation and gallbladder stones with normal cystic duct. Re s ults Among 371 patients, 63 had anatomic variations, A among which, 22 had compound variations with 85 variation points in total.The percentage of mutation rate was16.98%. 53 patients with variation of cystic duct had gallbladder stones, and 182 patients without variation of cystic duct had gallbladder stones. Conclus ions MRCP can clearly display different variations of cystic duct, and the patients with variation of cystic duct have higher risks in gallbladder stones than the patients without variations.

Objective To investigate the genotype of D class of carbapenemase of carbapenem-resistant Acinetobacter baumannii in traumatology department, so as to guide the rational use of antibiotics as well as prevent the nosocomial infection. Me thods A total of 96 strains of Acinetobacter baumannii were collected in traumatology department. Patient history were analyzed and genotype of carbapenemases of OXA-51, OXA-23, ISAba1-oxa-51 and ISAba1-oxa-23 were analyzed by PCR.Re s ults 70.84% (68/96) of Acinetobacter baumannii were isolated from wound tissue. 12 antibacterial agents were tested, 96 strains of Acinetobacter baumannii showed the highest resistance rate (78.13%) to imipenem and the lowest resistance rate (43.75%) to levofloxacin. Among 23 strains of imipenem-sensitive Acinetobacter baumannii, OXA-51 gene expression was not detected in 4 strains, however, OXA-23 gene expression were observed in 5 strains. The gene expression rates of OXA-51, OXA-23, ISAba1-oxa-51 and ISAba1-oxa-23 genes were 100%, 95.89%, 79.45% and 71.23% respectively in 73 strains of imipenem-insensitive Acinetobacter baumannii. The rate of gene expression profile of OXA-51+ISAba1-oxa-51+OXA-23+ISAba1-oxa-23 was 65.75%.Conclus ion The strains of imipenem-sensitive Acinetobacter baumannii may have the gene expression of OXA-23 of D class of carbapenemase.The main pattern of carbapenemase-producing was OXA-51+ISAba1-oxa-51+OXA-23+ISAba1-oxa-23 in traumatology department, which might be regarded as one of the mechanisms that led to the high resistance of the bacterium of acinetobacter baumannii to carbapenem antibiotics.

Objective To provide evidence-based prevention of chronic disease and nutritional interventions by investigating the development of dyslipidemia and hyperuricemia in rural areas of Yunnan and analyzing the relationship between hyperuricemia and dyslipidemia. Me thods The cross-sectional study recruited 513 residents over 18 years old in 2016. The morning fasting venous blood of all subjects were obtained to detect uric acid, serum total cholesterol (TC) , triglycerides (TG) and high-density lipoprotein cholesterol (HDL-C) . Each food factor's blood lipid to the hyperuricemia was determined through multivariate logistic regression. Re s ults The prevalence rates of hyperuricemia and dyslipidemia were 18.7% and 44.7% respectively. Females were more likely to have hyperuricemia than males (P <0.05) . Males were more likely to develop dyslipidemia than females (P >0.05) . The prevalence of hyperuricemia and dyslipidemia increased with age. Logistic regression analysis showed that the level of TG and LDL-C were risk factors and the level of HDL-C was protective factor of hyperuricemia.Conclus ion Regularly testing the levels of uric acid and blood lipid, enhancing the knowledge of reasonable diet and developing healthy dietary habits have significant importance to prevent chronic diseases.

Me thod Weselected 308 undergraduates with 165 (4 classes) majoring in clinical medicine and 143 (4 classes) majoring in medical laboratory in Kunming Medical Universityas the research object.Students were randomly divided into two groups and taught using demonstration method. One group practiced abdominal examination on other students while the other group adopted computer simulation.Both groups were giventests of practical skills and anonymous open-ended questionnaires after class.Re s ults The average score of computer simulation group was higher than the other group. Objective To investigate the application of the OSCE training on the intensive training of nursing skills for college students majoring in nursing in the fourth semester.Me thods Students from 2013 undergraduate session were investigated.They were randomly divided into two groups. The experimental group (n = 134) was trained of the OSCE training, while the control group (n = 135) received of traditional training. After the training, analysis of test scores and questionnaires were used to survey the comments of the two groups of students on intensive training of nursing skills. After two months of practice, the clinical comprehensive ability of the students was evaluated by clinical teachers.Re s ults The experimental group's theory and nursing skills were higher than those of the control group (P<0.01) . The experimental group had a positive attitude towards nursing OSCE training. Their familiarity with the operation skills, team cooperation spirits, presentation and communication ability, observation ability, clinical adaptation ability and health education ability were enhanced (P<0.01) .Conclus ion The comprehensive simulation training cannot only improve the effectiveness of OSCE training, but also strengthen the students' clinical comprehensive ability and promote the practice quality.

Objective To evaluate the impact of different rating instruments used by raters with different seniority in OSCEs. Me thods Surgical interns were randomly divided into two groups to undertake a nine-station surgical OSCEs. Raters with different seniority were also divided into two groups with four senior raters and four junior raters in one group. OSCE results were compared. Re s ults There was no statistical significance when comparing results from different raters using checklist rating instrument. Results from different raters using global rating scale instrument were significantly different. Correlations between raters using checklist and global rating scale instruments showed statistical significance in 81.3% of stations. Conclus ion Checklist rating instrument showed high capability of repeating the results. Therefore, it is recommended to apply checklist as many as possible in OSCE exams with mixed rater seniorities. Research on design of rating instruments are to be investigated to insure the effectiveness as well as the credibility of the rating system.

Objective To discuss clinical effects and security of using Ahmed valve implantation surgery to cure neovascular glaucoma.Me thods The clinical data of 78 patients with neovascular glaucoma in the Department of Ophthalmology from May 2015 to May 2017 were collected and retrospectively analyzed. The patients were randomly divided into the research group and the control group. Both groups were treated with Ahmed valve implantation surgery. Patients in the research group were treated with the professional nursing intervention of neovascular glaucoma. The intraocular pressure (IOP) and clinical effects of both groups before and after treatment were compared.Re s ults The achievement ratio of operation in the research group and the control group was 87.5%and 84.2%, respectively. There was no statistical difference (P >0.05) . The post-operation follow-up showed that average IOP in both groups was obviously lower than that in pre-operation (P <0.01) . But average post-operative IOP in both groups had no statistical difference (P >0.05) . There were three patients with complications in the research group, showing 10.0% of complication ratio. There were 10 patients with complications in the control group, showing 26.3% of complication ratio, which was the statistically different (P <0.05) .Conclus ions The curative effects of using Ahmed valve implantation surgery to cure neovascular glaucoma should be affirmed. The operation shows good short-term effects and reduction of complications but more long-term effects.By coordinating with nursing interference, long-term complications can be reduced. Therefore, it is an effective method to cure glaucoma.
The Influence of Family Nursing Intervention on Treatment Effect and Quality of Life among Asthmatic Children

Abstract:
Objective To investigate the effect of the family nursing intervention on treatment of children with asthma, and explore the effect of family nursing intervention on the pulmonary function and quality of life in asthmatic children. Me thods A total of 260 children with asthma were recruited at the First Affiliated Hospital of Xi'an Jiaotong University from July 2012 to July 2015. They were randomly divided into observation and control groups with 130 cases in each group. The control group were given routine nursing and regular treatment, and the observation group were received additional family nursing intervention management at the same time. We use Pediatric Asthma Quality of Life Questionnaire (PAQLQ) and pulmonary function testing to evaluate the influence of family nursing intervention on treatment effect and quality of life of asthmatic children. Re s ults The total effective rate was 93% in the observation group and was 65% in the control group (P<0.05) . The pulmonary function and quality of life were significant higher in the intervention group compared with the control group after intervention.Parents' satisfaction was significantly higher in the intervention group (P<0.05) . Conclus ion Family nursing intervention is an effective mode of management of children with asthma that can obviously improve pulmonary function and quality of life of asthmatic children. Moreover, family nursing intervention also contributes to raising parents' satisfaction.
