The Spation-temporal Expression of SOX9 during the Development of Mandibular Condylar Cartilage and Tibial Growth Plate Cartilage in SD Rats

Abstract:
  Objective  To explore the spation-temporal expression changes of SOX9during the development of condylar cartilage and tibial growth plate cartilage in SD rats.   Methods  Thirty adult SD rats were caged at the female-male ratio of 2∶1. The vaginal plug appeared was designated as gestation day 0 (GD0) and the rats were isolated then and raised in the routine way. The embryos were obtained from day 13 to day 21 respectively and the condylar cartilage and tibial growth plate cartilage were harvested. Serial section was performed to obtain the slices of the two kinds of cartilages at different development stages (E13 d-E21 d). HE staining and immunohistochemical detection were employed to compare the structure distinguish between two kinds of cartilages and examine the expressional changes of SOX9. The data were analyzed by applying single factor analysis of variance and two independent sample t test in SPSS17.0 software package.   Results  Some morphological differences, mainly in the forms, stratification and arrangements of cells, were observed between the condylar cartilage and the tibial growth plate cartilage during E14 d-E21 d indicated by HE staining. In tibial growth plate cartilage, the SOX9 positive expression in E15 d, E17 d and E20 d were stronger than those in condylar cartilage (P < 0.05). The distribution of positive cells in tibial growth plate and cartilage condylar cartilage was basically identical. SOX9 was mainly positive in the proliferation layer of the two kinds of cartilage.   Conclusions  The main structures of condylar cartilage and tibial growth plate cartilage during the development process are similar. The minority distinguished part is the layered form, which indicates different growth characteristics during embryonic development. During embryonic development, SOX9 is expressed in the two kinds of cartilages above and mainly distributes in the proliferative zone, which shows SOX9 is involved in the cell proliferation and differentiation associated with the differentiation of cartilage cell and development of cartilage.
Effects of Different Accumulation Culture Methods on the Titer of Phage PEf771

Abstract:
  Objective  To compare the effects of three different enriching culture methods on the titer of phage PEf771, and discuss the methods to effectively improve the titer of phage PEf771.   Methods  Through phage PEf771/E. faecalis YN771 proportional method, spot digging method and mitomycin C induction method, phage PEf771 stock solution was added at E. faecalis YN771 different growth stages for enrichment to obtain a higher titer of phage PEf771.   Results  The titer of PEf771 obtained by PEf771/E. faecalis YN771 = 1∶3 and spot digging method was higher, there was no significant difference between the two methods after expanded culture (P > 0.05), and the titer reached 108 pfu/mL in 24 hours. Mitomycin C induction method could not significantly improve the titer of PEf771.PEf771 was added in the early growth stage (OD600 = 0.3) and logarithmic stage (OD600 = 1) of E. faecalis YN771 for enrichment culture. The titer of the former was 1010 pfu/mL after 6 hours of culture, and remained 1010 pfu/mL after 24 hours, reaching the ideal titer.   Conclusion  Adding PEf771 in the early stage of E. faecalis YN771 culture (OD600 = 0.3) and enriching culture with PEf771/E. faecalis YN771 = 1∶3 or spot digging method can effectively improve the titer of PEf771. It provides experimental materials and basis for the study of phage PEf771 of E. faecalis YN771 in the treatment of infection caused by E. faecalis YN771.
Effect of Surface Topography on Staphylococcus Epidermidis Biofilm Formation by Different 3D Printing Thickness of Biomaterials

Abstract:
  Objective   To investigate the effect of surface topography on staphylococcus epidermidis biofilm formation by different 3D printing Thickness of biomaterials.   Methods   Medical 3D printing raw material, photosensitive resin MED610 was used as the material and samples were made according to 16 μm, 30 μm and 100 μm layer thicknesses using light-curing molding technology, respectively. Measure the sample surface roughness contour arithmetic mean deviation Ra, contour maximum height Rz, static angular contact method to detect sample hydrophobicity. The samples were co-cultured with Staphylococcus epidermidis standard strain RP62A on an oscillator, and the material surface was removed at 2 h, 6 h, 12 h, 24 h and 30 h, respectively. The number of bacterial communities per field of view was measured by laser confocal microscopy and the biofilm formation on the material surface was observed by scanning electron microscopy.   Results   The Ra and Rz values on the surface of the 16-μm layer thickness fabricated biomaterial were smaller than those of the 30-μm layer thickness and 60-μm fabricated samples, and there was no significant difference in the hydrophobicity of the surface of the fabricated materials with different layer thicknesses. When co-cultured with Staphylococcus epidermidis for 2 h and 6 h, a small amount of Staphylococcus epidermidis was scattered on the surface of the material in the 16 μm group without bacterial aggregation, and the number of bacterial communities per unit field of view was significantly lower than that in the 30 μm and 100 μm groups (P < 0.05). Biofilm formation could be observed on the material surface of each group at 12 h, 24 h and 30 h of incubation and there was no significant difference in the number of bacterial communities per unit field of view ( P > 0.05).   Conclusions   Different precision fabrication has a large effect on the surface roughness of 3D printing materials, but has no significant effect on hydrophobicity. The 16-μm layer thickness fabrication material is less favorable for Staphylococcus epidermidis adhesion in the early stage of infection than the 30-μm and 100-μm layer thickness fabrication materials.
Establishment of Spinal Cord Ischemia Injury Model in Rats by Thoracic Aorta Occlusion Combined with Aortic Bypass

Abstract:
  Objective  To modify the rat model of spinal cord ischemia injury.   Methods  The rats were divided into three groups. In experimental group, thoracic aorta was cross-clamped between T4 and T11, and aortic bypass circulation was established from proximal thoracic aorta to proximal diaphragmatic aorta. The interval of clamping was 20 minutes. In control group, the thoracic aorta (T4) was cross-clamped for 20 minutes.In the sham operation group, only thoracotomy was performed without thoracic aorta cross-clamping. Thefunction of spinal cord was monitored by transcranial motor evoked potential (TC-MEP). The pathology of spinal cord were observed 7 days after surgery. The locomotor function of spinal cord was evaluated by BBB score at multiple postoperative time. The survival curves were analyzed 28 days after operation.   Results  The TC-MEP waveform of the experimental group disappeared after 6 minutes, and that of the control group disappeared after 8 minutes until the end of surgery. The slices of spinal cord showed a large number of degeneration and necrosis of neurons in the experimental group. Multiple neurons were necrotic in the control group. The BBB score of the experimental group decreased more obviously than that of the control group. There was a significant difference in the 28-day survival rate between the experimental group and the control group, and (P < 0.05).   Conclusion  Thoracic aortic cross-clamping plus aortic bypass is an effective method, with high long-term survival rate.
miR-145 Regulates the Expression of OCT4 in Human Endometrial Stromal Cells and Promotes the Development of Endometriosis

Abstract:
  Objective  To further explore the regulatory mechanism of miR-145 in Endometriosis (EMs) through cell experiments.  Methods  miR-145 or octamer-binding transcription factor 4 (OCT4)gene and protein expression levels in cells were examined using reverse transcription-quantitative PCR and western blotting, respectively. Correlation analyses and dualluciferase reporter assays were performed to assess the association between miR-145 and OCT4. A Cell Counting Kit-8 assay was performed to test cell viability, while cell apoptosis was measured using flow cytometry. Moreover, the migration of hESCs was measured via Transwell assays.   Results  In cell assays, overexpression of miR-145 signifcantly promoted proliferation and migration (P < 0.01), but inhibited the apoptosis of hESCs (P < 0.05). Furthermore, the transfection of hESCs with miR-145 mimics decreased the protein expression levels of OCT4, Bax and MMP1 (P < 0.01), as well as increased the protein expression of Bcl-2. However, knockdown of miR-145 reversed these results and significantly inhibited the proliferation of hESCs by targeting OCT4 (P < 0.05).   Conclusion  The present results suggested that knockdown of miR-145 signifcantly suppressed the development of EMs by targeting OCT4, which may serve as a potential target for the treatment of EMs.

Abstract:
  Objective  To study the characteristics of left ventricular strain and myocardial pathological changes in acute myocardial injury induced by anthracyclines in Diannan small-ear pigs.   Methods  Six small-eared pigs from southern Yunnan were selected. After routine anesthesia, electrocardiogram electrodes were connected, daunorubicin was injected intravenously, and an animal model of acute cardiotoxicity was made. The echocardiography was performed before and three days after the administration. Data acquisition, blood draw for cTnT determination, and pathological examination of heart and liver after execution. Obtain two-dimensional echocardiography of ultrasound parameters to measure the left ventricular end-diastolic diameter (LVDd), peak E velocity, peak A velocity and E/A ratio of blood flow during diastole of the mitral valve, and automatically calculate the overall left ventricle longitudinal strain GLS and left ventricular stratified longitudinal strain parameters were compared and analyzed.   Results  There was no statistical difference in left ventricular end diastolic diameter (LVDd), left ventricular end systolic diameter (LVDs), ventricular septal end diastolic thickness (IVSd) and left ventricular posterior wall end diastolic thickness (PWd) and EF between before and after administration of acute myocardial injury (P > 0.05). Although the serum cTnT concentration gradually increased after administration, it remained within the normal range, and there was no significant statistical difference from the value measured before administration. The liver and myocardial cells showed edema and congestion to varying degrees, and hyaloid and mucus-like changes appeared in the endocardium (P > 0.05). There were statistical differences in the overall GLS of the left ventricle and the endocardial layer before and after the administration, but there was no statistical difference in the GLS of the middle layer and the epicardial layer (P < 0.05).   Conclusions  Large doses of anthracyclines can cause acute myocardial damage to the myocardium, and the pathological results support myocardial cell damage. At the same time, the conventional measurement of the size of the heart cavity and EF cannot reflect the functional damage of the myocardium in time. The use of 2D-STI technology to measure the longitudinal strain of the left ventricle and the strain of the center of the endocardium layer can be more sensitive to find applications than traditional cardiac ultrasound data such as EF. Abstract:
  Objective  T o optimize the ultrasonic extraction method for Geraniin, Gallic acid, Corilagin, Hyperoside, Ellagic acid, Protocatechuic acid from Geranium wilfordii Maxim by L9(34) orthogonal experiment.   Methods  On the basis of single factor experiments, using L9(34) orthogonal experiment, three main factors, methanol volume fraction, extraction time, solid-liquid ratio are chosen for three main component to study the effect of the interaction of various factors on the extraction rate of six components fromGeranium wilfordiiMaxim.   Results  The results show that the optimum extraction process are as follow: the methanol volume fraction was 60%, the solid-liquid ratio was 1∶10(g∶ml), the extraction time was 40 min.   Conclusion  This method is simple, repeatability and accuracy. It can be to extract Geraniin, Gallic acid, Corilagin, Hyperoside, Ellagic acid, Protocatechuic acidfrom Geranium wilfordii Maxim.
miR-142-5p Regulates the Proliferation and Metastasis of Gallbladder Carcinoma Cells through CCND1

Abstract:
  Objective  To explore the effect of miR-142-5p on the proliferation and metastasis of gallbladder cancer through CCND1 and to clarify the special mechanism of action.   Methods  RT-qPCR was performed to identify the differential expression of miR-142-5p in human gallbladder epithelial cells HGBEC and human gallbladder cell lines. Dual luciferase reporter gene assay was used to verify the targeting relationship between miR-142-5p and CCND1. CCK-8 was used for detecting human gallbladder cell proliferation. Transwell and wound healing assays were carried out to detect gallbladder cell migration. Western blot was performed to detect the marker expression of epithelial mesenchymal transition.   Results  miR-142-5p was significantly low expressed in gallbladder cancer cell lines, and lowest expressed in GBC-SD cells. Dual luciferase reporter gene assays revealed that miR-142-5p was able to inhibit the luciferase activity of wild-type CCND1.   Conclusion  Overexpressing of miR-142-5p inhibited the proliferation and metastasis of GBC-SD cells, simultaneous overexpression of miR-142-5p and CCDN1 partially reversed the effect of overexpressing CCDN1 on the proliferation and metastasis of GBC-SD cells. We demonstrate that miR-142-5p can inhibit the proliferation and metastasis of gallbladder cancer cells by suppressing CCND1 expression.
Investigation and Analysis on Health Literacy of Students in a Medical College in Yunnan Province

Abstract:
  Objective   To understand the health literacy of medical college students in Yunnan Province and to analyze its influencing factors, so as to provide basis and targeted strategies for future interventions.   Methods   Random cluster sampling method was used, and 2182 medical students were investigated by using the 2020 Chinese Citizens Health Literacy Questionnaire.   Results   The proportion of college students with health literacy was 84.7%, among which 68.1%, 77.2% and 75.9% had basic healthy knowledge, healthy lifestyles or behaviors, and healthy skills, respectively. The proportion of male and female college students with health literacy levels was 76.5% and 89.2%, respectively, the difference was statistically significant (P < 0.01). The proportion of students with healthy knowledge literacy from grade one to grade five was 60.3%, 66.0%, 74.9%, 74.3% and 76.7% , respectively, with a significant statistical difference ( P < 0.01). The logistic regression analysis showed that gender and family education level were the primary factors affecting healthy literacy of medical students.   Conclusions   The health literacy of female medical students is better than that of male students. With the increase of grades, the healthy knowledge literacy of medical students continues to improve. Medical students with higher family education level tend to have poor healthy literacy.

EGFR Gene Mutation State and Its Clinical Significance in Multi-nodular Lung Adenocarcinoma in Yunnan Province

Abstract:
  Objective  To investigate the relationship between EGFR gene mutations and clinicopathological characteristics of patients with multinodular lung adenocarcinoma in Yunnan province by using NGS method.   Methods  A total of 111 pulmonary nodule samples from 79 patients with multifocal lung adenocarcinoma were collected between January 2018 to May 2020 in the Molecular Diagnostic Center of Yunnan Cancer Hospital. The NGS method was used to detect the EGFR mutation of multiple lung nodules to analyze its relationship with clinicopathological characteristics, and Logistic regression to analyze its independent risk factors.   Results  The total EGFR mutation rate in 111 lung nodule tissue samples was 58.55% (65/111), among which L858R point mutation and exon 19 deletion were the most common, accounting for 53.8%of the total mutations (35/65).The rare mutation rate was 27.02% (30/111), the single point mutation rate was 37.84% (42/111), and the compound mutation rate was 20.72% (23/111). χ2 test showed that females, age ≥56 years, non-smoker, areas with a high incidence of lung cancer in Yunnan, earlier pathological stages (stages Ⅰ to Ⅱ), and invasive adenocarcinoma had a higher EGFR gene mutation rate, and the difference was statistically significant (P = 0.036、P = 0.001、P < 0.001、P = 0.006、P = 0.006、P = 0.034).Multivariate Logistic regression analysis showed thatage ≥56 years, non-smoker, areas with a high incidence of lung cancer in Yunnan, and earlier pathological stages (stages Ⅰ to Ⅱ) are independent risk factors for EGFR gene mutations (P<0.05).The heterogeneity rate of EGFR gene expression between paired nodules in the same patient was as high as 87.5% (28/32).  Conclusions  Among patients with multinodular lung adenocarcinoma in Yunnan, the elderly, non-smoker, areas with a high incidence of lung cancer in Yunnan, and earlier pathological stages (stages Ⅰ to Ⅱ) are independent risk factors for EGFR gene mutations.The high-level heterogeneity of driver mutations among the lesions of patients with multinodular lung adenocarcinoma suggests that multiple nodules tend to be independent of the origin of the primary tumor, which will provide more options for the diagnosis and treatment of multinodular lung adenocarcinoma.
Diagnostic Value of Guidewire Localization of Breast Mass Based on High-frequency Ultrasound Guidance in Breast Lesions

Abstract:
  Objective  To evaluate the diagnostic value of high-frequency ultrasound-guided guide wire localization of breast mass in patients with breast lesions.   Methods  Ninety patients with suspected breast mass lesions in Yunnan Hospital of Traditional Chinese Medicine from May 2020 to August 2021 were selected as subjects by random sampling method. After admission, the patients received high-frequency ultrasound-guided guide wire localization diagnosis of breast mass, and the pathological tissue examination results were used as the "gold standard". The diagnostic value of high-frequency ultrasound-guided guide wire localization of breast mass in patients with breast lesions was calculated and analyzed.   Results  All 90 patients with suspected breast mass lesions had complete pathological results. The results showed that 61 patients had benign lesions. 29 cases were malignant lesions. There was no significant difference in the detection rate of ultrasound-guided guide wire localization in fibroadenoma, breast adenosis, intraductal papilloma, invasive ductal carcinoma and invasive lobular carcinoma compared with pathological examination (P > 0.05). 57 cases of patients with benign breast lesions were diagnosed by ultrasonic-guided guide wire localization of benign breast masses, and the coincidence rate with gold standard diagnosis was 86.67%[(53+25) /90], the difference was statistically significant (P < 0.05). The diagnostic sensitivity and specificity were 86.89% (53/61) and 86.21% (25/29). Ultrasound guided guide wire localization of benign breast mass was confirmed in 27 cases of breast malignant lesions, and the coincidence rate with gold standard diagnosis was 88.89%[(57+23) /90] (P < 0.05). The diagnostic sensitivity and specificity were 79.31% (23/29) and 93.44% (57/61) respectively.   Conclusion  High frequency ultrasound-guided guide wire localization of breast mass for patients with breast lesions can obtain a high diagnostic rate, high diagnostic sensitivity and specificity, which can guide clinical diagnosis and treatment, and is worthy of promotion and application.
Changes of Thyroid Function and Its Clinical Significance before and after Radiotherapy and Chemotherapy in Patients Undergoing Radical Breast Cancer Treatment

Abstract:
  Objective  To analyze the changes of thyroid function and its clinical significance after chemotherapy and radiotherapy in patients undergoing radical surgery for breast cancer.   Methods  One hundred and six patients admitted to Yunnan Cancer Hospital from January 2015 to December 2018 with radical breast cancer undergoing chemotherapy and sternoclavicular combined field radiotherapy and regular review of thyroid function were selected as study subjects, and the thyroid function indexes of the patients were tested before chemotherapy, before radiotherapy, and 3, 6, 9, 12, 18 and 24 months after radiotherapy, respectively. The changes in hormone levels were observed.   Results   TSH was elevated in patients after radical breast cancer treatment 6 months after the end of radiotherapy compared with before chemotherapy(P < 0.05). TG-Ab was elevated 12 months after the end of radiotherapy compared with before chemotherapy(P< 0.05). TSH was elevated 3 months after the end of radiotherapy compared with before radiotherapy(P< 0.05). TG-Ab and TPO-Ab were elevated 6 months after the end of radiotherapy compared with before radiotherapy(P < 0.05).   Conclusion   Patients undergoing chemotherapy and radiation therapy for radical breast cancer can lead to the development of hypothyroidism and cause thyroid autoimmune reactions, which should be tested regularly and treated promptly.
Correlation between Neutrophil-lymphocyte Ratio and Disease Activity in 536 Patients with Systemic Lupus Erythematosus

Abstract:
  Objective  To assess the correlation between neutrophil-lymphocyte ratio (NLR) and the disease activity of systemic lupus erythematosus (SLE).   Methods  A total of 536 patients with SLE from the Department of Rheumatology and Immunology of the First Affiliated Hospital of Kunming Medical University were selected. According to the SLE Disease Activity Index (SLEDAI) score, patients with SLE were divided into inactive group, lightly active group, moderately active group, and severely active group. The differences in NLR among the four groups were compared, and the correlation between NLR and SLEDAI score was analyzed through Spearman correlation analysis.   Results  There were differences in NLR levels among the four groups. The results of pairwise comparison showed that the NLR level of the moderately active group was higher than that of the inactive group (adjusted P < 0.001); the NLR level of the severely active group was higher than that of the inactive group (adjusted P < 0.001); the NLR level of the severely active group was higher than that of the lightly active group (P = 0.011 after adjustment). The NLR of SLE patients were positively correlated with SLEDAI scores (P < 0.05).   Conclusion  NLR can be used as a reference index to evaluate the disease activity of SLE patients.

  Objective  To compare the clinical efficacy of double plates and locking plates in the treatment of complex proximal humerus fractures.   Methods  Fifty-seven patients admitted to the Department of Bone and Trauma Surgery of the Fourth Affiliated Hospital of Kunming Medical University from 01 2015 to 01 2020 who underwent surgical incisional reduction and internal fixation treatment and met the inclusion criteria for proximal humerus fractures were selected. According to the different fixation methods, they were divided into the double plate group (group A, 28 cases) and the locked plate group (group B, 29 cases), and the clinical efficacy of the 2 groups was comprehensively evaluated by comparing the indices of surgery, treatment effect, postoperative functional recovery and the occurrence of complications.   Results  The operating time, intraoperative bleeding, intraoperative fluoroscopy time, and postoperative incisional drainage were worse in group A than in group B (P < 0.05), but the postoperative loss of neck stem angle, humeral head inversion angle, loss of humeral height, Constant function score, and VAS score were better in group A than in group B (P < 0.05); there was no significant difference in the overall postoperative complication rate (P > 0.05 ).   Conclusion  Both double plates and locking plates can effectively treat complex proximal humerus fractures, but double plates are more effective in terms of fixation stability and functional recovery, and are more suitable for treating patients with complex fracture types, severe comminution, and incomplete medial column support.
Potential Value of G6PD Activity Detection in the Peripheral Blood for the Diagnosis and Prognosis of Cervical Cancer Patients with High Risk Human Papillomavirus Infection

Abstract:
  Objective  To explore the value of detecting G6PD activity in the peripheral blood for the diagnosis and prognosis of cervical cancer patients with high-risk human papillomavirus (HR-HPV) infection.   Methods  A total of 53 cervical cancer (CC) patients, 58 cervical intraepithelial neoplasia (CIN) patients and 40 health cohorts (HC), visiting and attending the clinic of Yunnan Third People’s Hospital during June 2014 to December 2015, were employed in this study. HPV Gene Chip Test Kit was used to detect and screen the genotype HPV infection in each group of population. Pathological evaluation and genotyping were performed on surgical specimens of cervical cancer patients. Serum samples were taken from each group of population, the activity of G6PD was detected by immunofluorescence assay, and the correlation analysis was carried out according to the clinical data and the results of G6PD activity. The results were then analyzed based on evidence.   Results  The activity of G6PD in peripheral blood was the highest in CC group, and there was significant difference to CIN Group and HC Group (P < 0.01) , and CIN Group to HC Group (P < 0.05). The change of G6PD activity in peripheral blood of CC patients is closely related to the progression of cervical cancer, and the change of G6PD activity in peripheral blood of CIN patients is closely related to the progression of cervical cancer. In CC group and CIN Group, the activity of G6PD in peripheral blood was consistent with HPV16/18 infection, and the relationship between the OS curve and the activity of G6PD in peripheral blood of CC patients infected with HPV 16/18 was analyzed, the 5-year survival rate decreased significantly in patients with high G6PD activity in peripheral blood (P <0.05) . The validity and reliability of G6PD activity detection and / or HPV16/18 infection test are valuable in evidence-based diagnosis for CIN or cervical cancer.   Conclusions  The activity of G6PD is related to the progression of cervical cancer. The detection of G6PD activity in peripheral blood can be used as one of the diagnostic and prognostic indicators of CIN and HR-HPV-infected cervical cancer, especially HPV 16/18.

Abstract:
  Objective   To study the relationship between the MTHFR C677T gene polymorphism and the collateral circulation in patients with carotid stenosis.   Methods   A total of 65 patients with moderate or severe carotid stenosis and chronic occlusion diagnosed by DSA and 45 controls without carotid stenosis from October 2018 to March 2021 in our hospital were included. All patients completed general assessment. 65 patients with moderate or severe carotid stenosis and chronic occlusion were evaluated the collateral circulation ASITN grading by DSA . All patients completed MTHFR C677T genetic polymorphism test.   Results   The incidence of stroke in patients with moderate or severe carotid stenosis and chronic occlusion was significantly higher than that in control group. There were significant differences in age, smoking history, hypertension, diabetes, total cholesterol, low density cholesterol lipoprotein, homocysteine and CT/TT type of MTHFR C677T gene between carotid artery stenosis or chronic occlusion group and control group (P < 0.05). The patients with good collateral circulation (ASITN grade 3-4) had high proportion of CC type in moderate or severe carotid stenosis and chronic carotid occlusion group. And the TT/TC type were more in patients with poor collateral circulation opening (ASITN grade 1-2) .   Conclusions   CT/TT type of MTHFR genotype is a risk factor for carotid stenosis. The collateral circulation of patients with CT/TT type is worse comparing with CC type in carotid stenosis.

Abstract:
  Objective  To compare the microbial differences of supragingival plaque between healthy gingiva group and gingivitis group of college students by high-throughput sequencing technology.   Methods  According to inclusion and exclusion criteria, 30 college students aged 18-25 with healthy gingiva and 33 with gingivitis were recruited from Kunming Medical University. Sequencing of 16S rDNA gene amplicon was performed using MiSeq system, and the diversity, structure and composition of supragingival plaque were compared between the two groups.   Results  A total of 87, 595 sequences were detected, including 14 phyla and 216 genera. Compared with the healthy gingiva group, the α diversity index in the gingivitis group was higher, and the difference was statistically significant (P < 0.001). The result of similarity analysis (Anosim) and principal component analysis (PCA) showed that there were significant differences in the structure and composition of the microflora of supragingival plaque between the healthy gingiva group and the gingivitis group (P < 0.001). The relative abundance of Prevotella, Porphyromonas and Bacillus in gingivitis group was higher, while the relative abundance of Actinomycetes in healthy gingiva group was higher.   Conclusion  There are significant differences in microbial community composition and structure of supragingival plaque between the healthy gingiva group and the gingivitis group of college students, and the diversity of the gingivitis group is higher.
Effect of Repairing Patellar Cartilage Fracture with Suture Anchor

Abstract:
  Objective  To study the effect of repairing patellar cartilage fracture with the suture anchor.   Methods  Between July 2016 and June 2018, 50 patients with patellar cartilage fracture were chosen, 18 male and 32 female, age 17 to 28. All of them were acute lateral dislocation of patella with fracture of patellar cartilage caused by trauma. Lysholm score is (63.8±7.2), VAS score is ( 8±1.7). All of the patellar cartilage fracture was repaired by suture anchor.   Results  All patients’wound healed well after operation, no swelling and effusion of knee joint, patellar cartilage healed well, no loosening of anchor, no displacement of cartilage fragments. Except for 3 patients with mild atrophy of quadriceps and poor recovery of muscle strength at 6 weeks after operation, the other patients’muscle strength recovered well. Lysholm score is (87.3±4.1), VAS score is ( 2±2.3), which were significantly improved compared with those before operation.   Conclusion  The operation technique which is fixation of patellar cartilage fracture with suture anchor is simple, can effectively fix cartilage fracture and avoid stimulating anterior patellar bursa, which has its unique advantages.

Abstract:
  Objective   To explore the effect of Kangfuyan capsule combined with hysteroscopy in the treatment of recanalization of fallopian tube after sterilization, and to analyze the effect of combined therapy on pregnancy rate.   Methods   A retrospective analysis of the clinical data of 121 patients with tubal ligation recanalization from February 2014 to February 2019. Patients were divided into control group and treatment group. In the treatment group, Kangfuyan capsules were taken orally for 1 month after recanalization. The control group did not take any special treatment after operation. The differences in intrauterine pregnancy rate and ectopic pregnancy rate of the two groups within 1 year were analyzed, and the correlation between age, postoperative fallopian tube length and other related factors were analyzed.   Results   Within 1 year after surgery, the intrauterine pregnancy rate in the treatment group was 79.6% which was much higher than the 56.1% pregnancy rate in the control group, P < 0.05; and the postoperative ectopic pregnancy rate in the treatment group was only 3.1%, which was far lower than the control group (10.5 %), P < 0.05. The intrauterine pregnancy rate in the control group stabilized over time, while intrauterine pregnancy in the treatment group was concentrated within 1-4 months after surgery. Age, location of sterilization, fallopian tube length after recanalization are highly correlated with intrauterine pregnancy rate.   Conclusion   Kangfuyan Capsules can increase the rate of intrauterine pregnancy in patients with tubal ligation recanalization, and reduce the rate of ectopic pregnancy

Abstract:
  Objective  To investigate the correlation of pulmonary function indexes and bronchial wall thickness measured as computed with high resolution computed tomography in patients with chronic obstructive pulmonary diseases.   Methods  Eighty COPD patients were assigned to four groups based on the new COPD Global Initiative Classification, meanwhile, fifty normal people with physical examination in the hospital were selected as controls simultaneously. All subjects underwent chest HRCT scanning to measure the bronchial wall thickness at the bilateral apical bronchial opening, and the correlation between the bronchial wall thickness and lung function was studied.  Results  Bronbronchial wall thickness was significantly increased in COPD patients compared with normal population, and the difference was statistically significant (P < 0.05). From the mild group to the very severe group, the bilateral bronchial wall thickness gradually increased, FEV1, FVC, FEV1/FVC gradually decreased, with statistically significant differences (P < 0.05). Correlation analysis: bilateral bronchial wall thickness was significantly inversely associated with each index of lung function (P < 0.05).   Conclusions  HRCT can be used to measure bronchial wall thickness and evaluate the degree of bronchial wall remodeling in COPD patients. With the deepening of the disease grade, the thickness of the bronchial wall of COPD patients gradually increased. There is a significant correlation between lung function indexes and bronchial wall thickness, that is, the thickening of bronchial wall, the decline of lung function indexes and the deterioration of lung function. HRCT measurement of bronchial wall thickness in COPD patients has clinical significance for the evaluation of the severity of the disease.

Abstract:
  Objective  To study the dynamic changes of serum AMH during different periods of treatment in patients who received the treatment of controlled ovarian hyperstimulation (COH) and to observe the correlation between AMH level in large and medium follicular fluid and the number of retrieved oocytes.   Methods  Serum AMH levels were measured on days 2-4 of natural menstrual cycle of baseline (BL), day 6 of follicular stimulation (S6) and day trigger. The AMH levels in the fluid of large and medium follicles were detected. The dynamic changes of serum AMH and its relationship with the number of retrieved oocytes were analyzed. The relationship between the level of AMH and the number of retrieved oocytes was compared between large and medium follicular fluid.   Results  (1) There was a positive correlation between serum AMH and the number of retrieved oocytes. There was a significant difference in serum AMH between BL and trigger, S6 and trigger (P < 0.05); (2) The value of AMH in large follicular fluid was positively correlated with the number of retrieved oocytes, and there was significant difference between large and medium follicular fluid (P < 0.05); (3) The value of AMH increased slowly from BL to S6, and the more the number of retrieved eggs, the more obvious the increase; The AMH value decreased rapidly from S6 to trigger day, and the more oocytes were retrieved, the more obvious the decrease was.   Conclusion  Serum AMH fluctuates during COH, and dynamic monitoring of AMH can predict the number of retrieved oocytes; There may be some guiding significance for the adjustment of Gn.

Abstract:
  Objective  To investigate the impact of acute non-normovolemic hemodilution (ANIH) on the numbers of NK cell, the NK cell cytotoxicity, numbers of NKT cell and the ratio of Th1 /Th2 cytokines in elderly patients undergoing rectal carcinoma radical resection.   Methods  Forty patients undergoing rectal carcinoma radical resection in the final date analysis were divided into two groups: Conventional Infusion (Group I, n = 20) and ANIH (Group II, n = 20). The venous blood samples were taken at 1 day before surgery (T1), at the first day after surgery (T2), the third day after surgery (T3) and the seventh day after surgery (T4) and measured the numbers of NK cell, NKT cell, Th1 and Th2 cytokines as well as the function of NK cell cytotoxicity by flow cytometry. Then we calculated the rate of Th1/Th2 to observe its variation trend.   Results  Compared with groups at T1, the NK cell numbers of the two groups at T2 and T3 both increased and the growth of cell numbers from group II had significant statistical deference with that of group I at T3 (P < 0.05). Oppositely the data of both groups reduced at T4 (P < 0.05) and the reduction from group I was obviously greater than group II (P < 0.05). Compared with T1, the function of NK cell cytotoxicity of both groups decreased at T2, and the change of group I was more obviously (P < 0.01). At T3 both groups increased markedly and compared with T1 and T2 it had statistical difference (P < 0.05). The data from group II rised higher than group I (P < 0.05). The data from both groups decreased at T4, which almost restore to T1 level. Compared with T2, the NKT cell number of both groups elevated at T3 (P < 0.05), and the augment of group II was more than group I (P < 0.05). At T4 the number of both groups went lower than at T3, and there was no significant statistical difference between of two groups. The change of Th1/Th2 made no sense from statistical perspective between two groups at any time point (P > 0.05).   Conclusion  Compared with conventional infusion, ANIH can protect and improve the perioperation cellular immunity in elderly patients undergoing rectal carcinoma radical resection as an effective measure of blood protection.

Abstract:
  Objective  To study the safety, feasibility and reliability of indocyanine green fluorescence staining in laparoscopic cholecystectomy, whether it can effectively avoid bile duct injury, and to explore the shortcomings and advantages of indocyanine green fluorescence laparoscopic technology in difficult cholecystectomy.   Methods  The method of retrospective analysis was adopted, and the clinical data of patients meeting inclusion criteria for difficult cholecystectomy in 65 cases of cholecystectomies with the causes of the gallstone and recurrent cholecystitis in The Second Affiliated Hospital of Kunming Medical University from December 1, 2018 to December 1, 2019 were collected. All patients had signed the informed consent and had no history of iodine allergy. 65 patients were assigned randomly with 35 cases of laparoscopic cholecystectomy (LC) and 30 cases of laparoscopic cholecystectomy (LC) with fluorescence cholangiography of indocyanine green (ICG). All kinds of the preoperative, intraoperative and postoperative data were collected to compare between the conventional group and cholangiography group. The application effect, safety and feasibility of the laparoscopic technologies with fluorescence cholangiography in the difficult cholecystectomy were explored, including whether the anatomic structure of the bile duct and the biliary tract time can be identified effectively.   Results  The time of laparoscopic cholecystectomy (LC) group was significantly longer than that of fluorescence cholangiography group (P < 0.05); in terms of the time to isolate the anatomy of biliary tract, the time of fluorescence cholangiography group was significantly shorter than that of laparoscopic cholecystectomy (LC) group (P < 0.05); the amount of bleeding in laparoscopic cholecystectomy (LC) was no statistically significant combined with fluorescence cholangiography group (P > 0.05); there was no significant difference in length of stay (LOS) and all-in cost of stay between the two groups (P > 0.005).   Conclusions  Indocyanine green fluorescence imaging laparoscopy has high operability and strong safety in identifying the anatomical structure of biliary tract in difficult cholecystectomy. It can effectively identify the anatomical structure of gallbladder triangle and effectively avoid the injury of biliary tract and blood vessels; Indocyanine green fluorescence imaging laparoscopic technology has the advantage of short operation time compared with traditional laparoscopic technology in Difficult Cholecystectomy, and there is no significant difference in postoperative liver function, infection index, hospitalization cycle and total hospitalization cost.

Abstract:
  Objective  To study the risk factors and intervention methods of secondary aspiration and aspiration pneumonia caused by swallowing dysfunction after stroke.   Methods  The patients with dysphagia after stroke were divided into observation group and control group. Two groups of patients with secondary SAP were divided into SAP group and non-SAP group for retrospective analysis; intervention treatment was given to the two groups of patients.   Results  SAP infection occurred in the two groups of patients with swallowing dysfunction after stroke. 43.33% of the observation group and 46.66% of the control group were 28 patients with SAP infection. Multivariate regression analysis showed that the age ≥ 65 years old, male sex, diabetes, Patients with symptoms of atrial fibrillation and nasogastric tube feeding were at greater risk of secondary aspiration pneumonia.   Conclusions  The risk of secondary SAP in patients with dysphagia after stroke should be paid more attention to. The higher the degree of basic diseases and neurological deficits in patients between 60-70 years, the greater the risk.

Nicotinic acetylcholine receptors (nAChRs) are the central regulators of stimulatory and inhibitory neurotransmitters that control the synthesis and release of growth,angiogenic and neurotrophic factors incancer cells, the cancer microenvironment and distantmetastasis. Smoking and other possibly environmental and lifestyle factors increase the function of nAChRs that up-regulates carcinogenesis and down-regulates carcinostasis. α7-nAChRis particularly overexpressed in lung cancer cells, and participates in the proliferation, apoptosis and migration of cancer cells.Thus,α7-nAChR might be a new therapeutic target for lung cancer.

Abstract:
  Objective  To investigate the prevalence and characteristics of nephropathy in HIV infected patients before antiviral treatment, and to analyze the related factors of nephropathy before antiviral treatment.  Methods  Patients with HIV infection who visited the Third People’ s Hospital of Kunming from June 2019 to March 2021 were recruited and screened by urine analysis, serum creatinine, urine microalbumin-to-creatinine ratios, urinary α1 microglobulin, and renal ultrasound. And all patients were screened by triglyceride, total serum cholesterol, fasting plasma glucose test . Body mass index was measured.Demographic data , medical history of hypertension and diabetes were collected. SPSS17.0 was used for univariate analysis and logistic regression analysis.  Results  The mean age of 468 subjects was 43.42±14.66 years. The male to female ratio was 1.96. The mean body mass index was 21.73±3.76. Body mass index ≥25 accounted for 15% of all the subjects . Some patients smoked daily which accounted for 33.3% of all the subjects. Patients with hypertriglyceridemia, hypertension and diabetes accounted for 24.7%, 10.7%, 7.7% respectively. And 21.8% of patients were taking drugs which were not antiretroviral therapy. The detection rate of kidney disease in total of 468 cases was 20.7%. the CKD-EPI formula was used to calculate and estimate glomerular filtration rate (eGFR). Patients with eGFR < 60 mL/min/1.73 m2, 60~90 mL/min/1.73 m2, ≥ 90 mL/min/1.73 m2 accounted for 1.1%, 9.6%, 89.3% respectively. The positivity rate of urinary protein was 3.1%, which measured just only by urine analysis. The urinary microalbumin/creatinine ratio (ACR) ≥ 30 mg/gCr were occurred in 24/111 cases (21.6%). The Urinary α1 microglobulin ≥ 12.5 mg/L were occurred in 71/111 cases (63.9%). Multivariate analysis indicated the independent effects of the age ≥ 50 years (OR = 2.285, P = 0.002), body mass index ≥25 (OR = 2.029, P = 0.025), hypertension (OR = 2.334, P = 0.017), diabetes (OR = 2.634, P = 0.014), HIV viral load ≥1×105 copies /mL (OR = 2.296, P = 0.002), which significantly associated with kidney disease. Female is a protective factor for kidney disease in HIV infected persons before antiviral therapy.   Conclusions  The detection rate of kidney disease is high and mainly abnormal urinary protein in Kunming area.Gender, age, hypertension, diabetes, body mass index and HIV viral load are significantly associated with the kidney disease in HIV patients before antiviral treatment. When screening for kidney disease, it is recommended to fully evaluate the results with urine analysis, serum creatinine, renal ultrasound, urinary microalbumin/creatinine ratio, and urinary α1 microglobulin.

  Objective   To explore the cluster nursing for preventing deep vein thrombosis of lower extremity after total hysterectomy for benign gynecological diseases.   Methods  A total of 90 patients with benign gynecological diseases after total hysterectomy from June 2020 to February 2021 were selected as the case group, and 88 patients from September 2019 to May 2020 were selected as the control group. Three indicators were compared between two groups, including the incidence of deep vein thrombosis of lower extremity, the satisfaction of patients, and satisfaction of medical staff with cluster nursing plan.   Results  The incidence of deep vein thrombosis of lower extremity in the case group was 1.11% (1/90), which was lower than that in the control group (7.95%, 7/88), and the difference was statistically significant (P < 0.05). After the application of cluster nursing, the satisfaction of doctors, nurses and patients were all above 97%, and the difference was statistically significant (P < 0.05).   Conclusion  Cluster nursing based on risk assessment contributes to prevent deep vein thrombosis of lower extremities after total hysterectomy for benign gynecological diseases.

Abstract:
  Objective  To analyze the impact of the “Internet health system” on the HAMA, HAMD, SCHFI score and SF-36 score in stroke patients.   Methods  A total of 150 stroke patients were included in this study. They were divided into two groups according to the length of treatment, with 75 cases in each group. 75 patients admitted to ~2019 in November January 2019 were included in the control group. 75 patients in December 2019 January in January 2019 were treated with traditional nursing. The trial group was given the Internet plus medical health system nursing. The incidence of adverse events, HAMA score, HAMD score, SCHFI score and SF-36 score were compared between the 2 groups after nursing intervention.   Results   (1) Incidence of adverse events: the total incidence of adverse events in the experimental group was 5.33%, which was significantly lower than that of 17.33% in the control group (P < 0.05); (2) psychological status: the scores of HAMA and HAMD in the experimental group were lower than those in the control group (P < 0.05); (3) self care ability and quality of life: the SCHFI score and SF-36 score of the patients in the trial group were higher than those in the control group (P < 0.05).   Conclusion  Internet health system nursing can effectively improve the negative emotions and quality of life of stroke patients.
