An Economical Method for Cultivation and Identification of Mast Cell Derived from Suckling Rabbit Bone Marrow

Abstract:
  Objective   To obtain more mature mast cells with more typical characteristics through a simple and well-operated in vitro induction culture system.   Methods   The bone marrow of suckling rabbits was blown out and put into high glucose medium containing DMEM, and the medium was changed every other day. When the cells were confluent, F12 induction medium was used, and the medium was changed every other day. When the cells became round under the microscope, 1640 maturation medium was replaced, and the medium was changed every 7 days until the cells were induced to mature. Toluidine blue staining was used to detect its function, and flow cytometry was used to detect the surface expression of CD117 and FcεR1α.   Results   After 6 weeks, toluidine blue staining showed that the cells containing more metachromatic particles could be obtained by adding β-mercaptoethanol. Flow cytometry showed that the proportion of CD117+ and FcεRIα+ double positive cells in the presence of β-mercapto ethanol was 96.2%, which was significantly higher than that in the absence of β-mercapto ethanol (P < 0.05).  Conclusion   The use of SCF, IL-3 and β-mercaptoethanol can successfully induce mast cells derived from rabbit bone marrow, which provides foundation for the future research on inflammatory injury in rabbits, and makes subsequent basic and clinical research possible.
Analysis of Characteristic Indexes of Liver Injury Model Induced by Different Antituberculosis Drugs in Mice

Abstract:
  Objective  To establish a mouse model of anti-tuberculous drug-induced liver injury, to explore the mechanism of anti-tuberculous drug induced liver injury, and to provide the basis of animal model.   Methods  Experimental mice were divided into 7 groups, including control group, group infected with Mycobacterium tuberculosis (MTB), the other 5 groups were given different drugs after infection with MTB, namely isoniazid (INH) 45 mg/(kg·d), rifampicin (RIF) 90 mg/(kg·d), ethambutol (EB)135 mg/(kg·d), pyrazinamide (PZA)180 mg/(kg·d), tetrad antituberculosis drugs (INH+RIF+EB+PZA), respectively. All doses were converted according to human dose and experimental animal dose. After 12 hours and 1-3 weeks of administration, the biochemical, liver index and pathological indexes were measured.  Results  After a single intragastric administration of INH, RIF, EB, PZA and tetrad anti-tuberculosis drugs for 12 hours, the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in RIF group and tetrad group mice increased (P < 0.05). After continuous intragastric administration of INH, RIF, EB, PZA and tetrad anti-tuberculosis drugs for 1 week, the serum indexes of ALT, AST and TBIL in the liver of INH, RIF and tetrad group mice were significantly increased (P < 0.05), and the liver index and pathological range were gradually increased and enlarged (P < 0.05). After continuous intragastric administration for 2-3 weeks, the serum indexes of liver ALT, AST, GGT and TBIL in INH, RIF and tetrad groups continued to increase (P < 0.05), the liver index continued to increase (P < 0.05) and the range of lesions expanded (P < 0.05). The correlation analysis showed that INH and RIF had a time cumulative effect, and the time of administration was significantly correlated with the serological index, liver index and lesion range of liver injury (P < 0.05), while the time of administration in other groups was not significantly correlated with the liver injury index (P > 0.05).   Conclusion  There is a trend of antagonistic liver toxicity among four combination anti-tuberculosis drugs, and INH and RIF are the main factors causing liver damage in mice. The pathological changes are mainly liver cell damage, and the abnormal degree of pathological indicators is earlier than serological indicators. The use of drugs to protect liver cell lesions in advance may have some clinical significance in preventing liver injury caused by anti-tuberculosis drugs.
Effect of MiR-26a-5p Regulating SLC26A4 to Ameliorate Hearing Loss in Deafness

Abstract:
  Objective  To explore the effect of miR-26a-5p in regulating SLC26A4 to amiliorate hearing loss, and to detect the expression changes of miR-26a-5p and SLC26A4 in deaf mice and hearing loss mice.   Methods  A mouse hearing loss model and a mouse deafness model were constructed, and the auditory brainstem response test was used to detect the hearing in the right ear of mice to determine whether the models were successfully constructed. qPCR test was used to detect the relative expression of miR-26a-5p and SLC26A4 after hearing loss and deafness in mice were established. The specific site of miR-26a-5p targeting binding to SLC26A4 was predicted at TargetScan website. The interference and overexpression vector of miR-26a-5p and SLC26A4 were injected, and the expression changes of miR-26a-5p and SLC26A4 were detected.   Results  After hearing loss and deafness were established, the expression of miR-26a-5p was significantly decreased (P < 0.05), while the expression of SLC26A4 was significantly increased (P < 0.05). After antagomir injection, miR-26a-5p was significantly down-regulated (P = 0.047), while SLC26A4 was significantly up-regulated (P = 0.014); and the hearing threshold of mice decreased significantly (P < 0.05), but the result was just the opposite after injection of miR-26a-5p agomir.   Conclusion   miR-26a-5p can ameliorate hearing loss through SLC26A4 and thus decelerate the development of deafness.
Mechanisms of Ginsenoside Rg1 in Protecting Cardiomyocytes by Sestrin2

Abstract:
  Objective   To explore the role and mechanisms of Sestrin2 in ameliorating hypoxia-reoxygenation injury of rat’s cardiomyocytes by ginsenoside-rg1 (G-Rg1).   Methods   Rat H9c2 cardiomyocytes cultured in vitro were randomly divided into 5 groups according to the experimental requirements: control group, Hypoxia reoxygenation group, G-Rg1 group, hypoxia Inducible Factor-1 (HIF-1, HIF-1α) inhibitor group, and Sestrin2 siRNA interference group. Flow cytometry was used to detect the expression levels of ROS in cardiomyocytes. Western blot was used to detect the protein expression of HIF-1α, Sestrin2, ATF-6, PERK, IRE-1 in cardiomyocytes. Immunofluorescence was used to test the protein expression of Sestrin2, HIF-1α, IRE-1.   Results   Compared with control group, the level of ROS and protein expression of HIF-1α、Sestrin2、ATF-6、PERK、IRE-1 of hypoxiareoxygenation group were significantly increased (P < 0.05) . Compared with the hypoxiareoxygenation group , the ROS level in G-Rg1 group were significantly reduced and protein expression of HIF-1α、Sestrin2、ATF-6、PERK、IRE-1 in G-Rg1 group were significantly increased (P < 0.05) . Compared with G-Rg1 group, the level of ROS were significantly increased and the protein level of Sestrin2 were significantly reduced, in HIF-1α inhibition group (P < 0.05). Compared with G-Rg1 group, the level of ROS were significantly increased and the protein level of IRE-1 were significantly upregulated in Sestrin2 siRNA group (P < 0.05).   Conclusions  G-Rg1 protects rat cardiomyocytes and maintains endoplasmic reticulum homeostasis in vitro by upregulating expression of Sestrin2.
Correlation between DDAH1 Gene 836A/T Polymorphism and Type 2 Diabetic Kidney Disease in Yunnan Han Population

Abstract:
  objective   To investigate the association between DDAH1 gene 836A/T polymorphism and type 2 diabetic kidney disease (DKD) in Yunnan Han population.   Methods   A total of 660 patients with T2DM were included. Patients were divided into three groups based on UACR results: simple type 2 diabetes mellitus group (DN0 group), complicated with early nephropathy group (DN1 group), and complicated with clinical nephropathy group (DN2 group). A total of 304 healthy people were included as normal control group (NC group). The genotype of DDAH1 gene 836A/T polymorphism was determined by PCR-DNA sequencing. The plasma asymmetric dimethylarginine (ADMA)concentration was measured by ELISA.   Results  The distribution frequency of AA genotype and allele A at the 836 locus of DDAH1 gene in the DN1+DN2 group was significantly higher than those in the DN0 group (P < 0.05). AA genotype carriers have higher ADMA levels than AT+TT genotype individuals in T2DM patients. The level of ADMA in DN1+DN2 group was higher than that in DN0 group (P < 0.05). Logistic regression analysis showed that ADMA level and DDAH1 gene 836 AA genotype were risk factors for DKD in T2DM patients.   Conclusions  The DDAH1 gene 836 AA genotype may be a risk factor for diabetic kidney disease in T2DM patients of Han nationality in Yunnan Province, and patients with this genotype have higher ADMA levels.
Growth and Development of Adolescent Girls of Lahu Ethnic Group in Lancang County of Yunnan Province

Abstract:
  Objective  To investigate the physical growth and sexual development of adolescents in Lancang Lahu Autonomous County, Yunnan Province, and establish a basic database of Lahu female growth and development.   Methods  A cluster random sampling method was used to select 2870 teenage girls from grades 7 to 9 from 5 middle schools in Lancang County. Local doctors measured the physical growth of adolescent girls and analyzed the physical and sexual development.  Results   In 2021, the height development grade evaluation of female adolescents in Lancang County, Yunnan Province was lower in rural areas than in urban areas (P < 0.01), and the height development of Lahu ethnic groups was lower than average, and tended to increase with age. The incidence of growth retardancy in rural areas (3.2%) was higher than that in urban areas (1.8%), P = 0.036, and that in Lahu ethnic groups (5.2%) was higher than that in other ethnic groups (P < 0.01). The incidence of high waist circumference and obesity in urban areas (25.9%, 10.5%) was higher than that in rural areas (16.8%, 4.7%, P < 0.01). The average age of menarche of female Lahu students in rural areas was later than that of female students of other ethnic groups (P < 0.01).   Conclusion  Female students of Lahu nationality have lower height development and higher incidence of growth retardation than other ethnic groups. The average age of menarche of Lahu female students living in rural areas was later than that of other female students. It is suggested that individual health education and dietary guidance should be taken according to the different growth and development of adolescents according to their nutritional status.
Current Situation and Influencing Factors of Breast-feeding in Four Districts of Kunming

Abstract:
  Objective   To understand the current situation and influencing factors of breastfeeding in the four districts of Kunming, and explore the room for improvement of the current breastfeeding rate in Kunming, so as to provide a realistic basis for breastfeeding knowledge publicity and training mode.   Methods   A convenient sampling method was used to select 5 tertiary hospitals and community health service centers in 4 districts of Panlong, Xishan, Wuhua District and Guandu of Kunming City. The mothers of infants and young children in the pediatric clinic and the child health and vaccination clinic were given questionnaires to conduct the breastfeeding questionnaire survey.   Results   44.7% of the infants were fed exclusively from 0 to 6 months, 36.6% were fed mixed from 0 to 6 months, and 18.7% were fed artificially from 0 to 6 months. The median duration of breastfeeding was 10 months and the median duration of exclusive breastfeeding was 4 months. Logistic regression analysis showed that social support, working during lactation, knowledge of breastfeeding, sleep status of mothers, self-efficacy, number of children’s diseases in the past year, and husband’s support for breastfeeding had statistical significance on the occurrence of breastfeeding (P < 0.05).   Conclusion   Breastfeeding in urban areas of Kunming is at a low level, affected by many factors, and has not yet reached the goals of the global strategy of infant and young child feeding, and it is necessary to develop a reasonable nursing plan to improve the breastfeeding rate and breastfeeding duration.

Clinical Study of Cervical Spondylotic Myelopathy Treated by Single Door Laminoplasty

Abstract:
  Objective  By analyzing the long term clinical effect of patients after single open-door cervical laminoplasty, the correlation between the change of cervical spinal cord area and the long term clinical effect of patients was discussed.   Methods  A retrospective analysis was performed on 60 patients with cervical spondylotic myelopathy who underwent cervical single open-door laminoplasty in the First Affiliated Hospital of Hainan Medical College from November 2015 to June 2021. The long-term clinical efficacy was evaluated by Japanese Orthopaedic Association (JOA ) improvement rate at the last follow-up. 32 patients with JOA improvement rate of > 60% at the last follow-up were divided into good prognosis group, while 28 patients with JOA improvement rate of ≤60% at the last follow-up were divided into poor prognosis group. Gender, age, duration of symptoms, blood loss, operation time, preoperative and postoperative JOA score were observed. Spinal canal sagittal diameter, cervical curvature index, cervical range of motion, spinal canal area and cervical spinal cord area were measured by preoperative and postoperative cervical MRI, CT and X-ray.   Results  The change of cervical spinal cord area in the good prognosis group was (55 ± 41 ) mm2, which was significantly larger than that in the poor prognosis group (35 ± 37 ) mm2, and the difference between the two groups was statistically significant (P < 0.05 ). The preoperative and postoperative spinal canal area and the axial position in the good prognosis group were significantly larger than those in the poor prognosis group (P < 0.05 ). But there was no significant difference in the preoperative and postoperative spinal canal ulnar diameter and cervical activity between the two groups (P > 0.05 ). Multivariate regression analysis showed that preoperative (OR = 6.924, P < 0.05) and postoperative JOA score (OR = 22.000, P < 0.05 ), preoperative spinal canal area (OR = 2.518, P < 0.05 ) and cervical spinal cord area change (OR = 5.513, P < 0.05 ) were related to the improvement rate of JOA at the last follow-up.   Conclusion  The change of cervical spinal cord area after SDCL, preoperative cervical spinal canal area, and preoperative and postoperative JOA score are closely related to the improvement rate of JOA at the last follow-up. Surgeons can predict the long-term efficacy of the patients according to the change of cervical spinal cord area after SDCL.
Correlation between Plasma proBDNF Level and Primary Sjögren’s Syndrome with Depressive Symptoms

Abstract:
  Objective  To study the role of plasma proBDNF level in patients with primary Sjogren's syndrome (pSS) with depressive symptoms, and to explore related factors, so as to provide evidence for early intervention.   Methods  A total of 42 pSS patients and 81 healthy controls were investigated with self-made general information questionnaire and PHQ-9 scale, and 42 pSS patients were investigated and scored with Coping style questionnaire (SCSQ) and social support rating scale (SSRS). European Antirheumatic Association pSS Disease Activity Scale (ESSDAI) was used to evaluate the disease activity of pSS. After informed consent was obtained, 3-5 ml peripheral blood was collected to detect the level of proBDNF.   Results  The increase of plasma proBDNF level in patients with pSS complicated with depressive symptoms was the most significant (P < 0.05), and less social support was a risk factor.  Conclusion  ProBDNF may be involved in the pathogenesis of pSS with depressive symptoms, and these patients need more social support.
Rehabilitation Efficacy of Two Exercise Methods on Adolescent Scoliosis

Abstract:
  Objective  To evaluate the efficacy of two different types of exercise in adolescent patients with scoliosis.   Methods  43 children who received rehabilitation treatment for scoliosis from January to December 2022 were included in this study and randomly divided into 2 groups, including Schroth and core group (n = 27) and home exercise intervention group (n = 16). Both groups exercised for 1 hour, 2 days a week. Height, weight, BIM, cobb angle, trunk rotation (scoliosis meter), and thoracic expansion (cm) were continuously evaluated for efficacy during treatment.   Results  After 12 months of rehabilitative treatment for patients with scoliosis, there were statistical significance between the trunk rotation angles in right thoracic segment and left lumbar segment, and thoracic expansion in Schroth and core group (P < 0.05), and there were statistical significance between the trunk rotation angle of left lumbar segment and thoracic expansion in the home exercise intervention group (P < 0.05).   Conclusion  Exercise therapy combined with bracing improved trunk rotation and thoracic expansion in mild and moderate scoliosis, and the earlier the detection and intervention, the better the curative effect. The Schroth and core group exercise therapy was superior to the home exercise intervention. Exercise therapy has an effect on trunk rotation angle in scoliosis.
Pelvic Floor Tthree-dimensional Ultrasound Evaluating Postpartum Pelvic Floor Dysfunction in Elderly Parturient Women

Abstract:
  Objective  To explore the changes of pelvic floor structure and function in early postpartum period of elderly parturient women by two-dimensional and three-dimensional ultrasound.   Method  A total of 86 early postpartum (6-8 weeks) elderly parturients admitted to our hospital from July 2021 to January 2023 were selected as the observation group, and 50 early postpartum parturients admitted during the same period were selected as the control group. All study subjects underwent pelvic floor two-dimensional and three-dimensional ultrasound diagnosis. The two groups of maternal pelvic floor two-dimensional ultrasound evaluation parameters, three-dimensional ultrasound evaluation parameters were compared. The diagnostic efficacy of pelvic floor 2D and 3D ultrasound was compared for puerperal pelvic fIoor dysfunction (PFD) in women of appropriate age and advanced age.   Result  The observation group showed significantly higher levels of resting bladder neck position (BNP), posterior urethral angle (PUA), Valsalva maneuver BNP level, and bladder neck desent (BND) compared to the control group (P < 0.05). Levator hiatal anteroposterior diameter (LHAP), levator hiatal lateral diameter (LHLP), and levator hiatal area (LHA) when performing Valsalva maneuver were significantly higher than that of the control group (P < 0.05). The sensitivity and accuracy of pelvic floor three-dimensional ultrasound (92.59%) were significantly higher than those of two-dimensional ultrasound (70.37%, 77.91%) (P < 0.05).   Conclusion  Functional and structural changes of the pelvic floor are more likely to occur in older women in the early postpartum period. Compared with two-dimensional ultrasound, three-dimensional pelvic floor ultrasound is more helpful to improve the diagnostic efficiency of PFD.
Preventive Effect of Adenosine for No-reflow Phenomenon in Patients with Acute Non-ST-segment Elevation Myocardial Infarction

Abstract:
  Objective   To study the effect of intracoronary injection of adenosine for the prevention of no-reflow phenomenon (NRP) before balloon dilation and stent placement during primary percutaneous coronary intervention (PPCI) in patients with acute non-ST-segment elevation myocardial infarction (NSTEMI).   Methods   A prospective randomized controlled design was used to select 196 NSTEMI patients who received PPCI in the Department of Cardiology, Yan’an Hospital, from January 2020 to June 2022. Randomized number table method was used to divide the patients into two groups, with 98 patients in each group. Patients in one group were treated with intracoronary injection of 100 μg adenosine prophylactically before stent placement or balloon dilation; while patents in another group were given 100 ug adenosine after stent placement if NRP happened. At the end of PPCI, the incidence of myocardial infarction thrombolysis test (TIMI) blood grading, myocardial TIMI perfusion grading (TMPG), myocardial staining grading (MBG) and NRP were examined.   Results   After stent placement, 89.6% of patients in the prophylactic administration group had blood flow TIMI, TMPG and MBG grades of infarct-related vessels reaching grade 3, and 73.4% in the remedial administration group, with statistical significance (P < 0.05). The incidence of NRP was 8.2% in the prophylactic group and the incidence was 23.6% in the remedial administration group, with statistical significance ( P < 0.05). TIMI, TMPG and MBG grade 3 blood flow were restored in all patients presented with NRP after intracoronary injection of adenosine.   Conclusion   For indicated patients with NSTEMI, prophylactical intracoronary injection of adenosine before stent implantation can reduce the incidence of NRP significantly after stent placement.

Abstract:
  Objective   : To investigate the efficacy of two-dimensional (2D) ultrasound combined with three-dimensional (3D) ultrasonic free anatomical imaging (Omni View) technology in the treatment of fetal cleft lip and palate.   Methods   37434 pregnant women who underwent prenatal ultrasound examination in the Affiliated Hospital of Yunnan University from January 2018 to December 2022 were selected as study subjects. All pregnant women were examined by 2D ultrasound alone and combined with 3D OmniView. Taking the results of natural delivery or induced labor as the gold standard, the diagnostic accuracy of 2D ultrasound and 2D ultrasound combined with 3D Omni View of fetal cleft lip and palate was compared.   Results   A total of 70 cases of fetal cleft lip and palate were confirmed by natural delivery or labor induction, 53 cases were confirmed by 2D ultrasound, and 64 cases were confirmed by 2D ultrasound combined with 3D Omni View technology, the difference between the two was statistically significant (χ2 = 6.295, P = 0.012).   Conclusion   2D ultrasound combined with three-dimensional Omni View, which can visually display the location of lesions and significantly improve the detection rate of fetal cleft lip and palate, can help clinicians in evaluation and consultation of pregnant women.
Value of Transesophageal Three-dimensional Echocardiography in Diagnosis of Perivalvular Leakage after Aortic Valve Replacement

Abstract:
  Objective  To explore the clinical value of transesophageal three-dimensional echocardiography (RT-3D-TEE) in the diagnosis of perivalvular leakage after aortic valve replacement.   Methods  70 patients with suspectde perivalvular leak after aortic valve replacement in our hospital from January 2015 to December 2018 were selected.All patients underwent 2-D transthoracic echocardiography (2D-TTE) and RT-3D-TEE was compared. The accuracy of the two methods in the diagnosis of perivalvular leakage after aortic valve replacement, and the location, range and other conditions of perivalvular leakage were compared. All 70 patients underwent surgery again.   Results  After surgical treatment, 45 cases of perivalvular leakage and 25 cases of non-perivalvular leakage were confirmed.2D-TTE was diagnosed correctly in 52 cases, misdiagnosed in 9 case and missed in 9 cases. RT-3D-TEE was diagnosed correctly in 70 cases. The diagnostic accuracy of RT-3D-TEE (100.0%) was higher than that of 2D-TTE (80.0%), and the difference was statistically significant (P < 0.05). Among the 36 cases of perivalvular leakage diagnosed by 2D-TTE, 5 were located in the left coronary valve region, 8 were located in the right coronary valve region, 11 had multiple or larger than a single lobe region, and the remaining 12 cases could not be clearly identified. Among the 45 cases of perivalvular leakage diagnosed by RT-3D-TEE, 9 cases were located in the left coronary valve region, 15 cases were located in the right coronary valve region, 18 cases had multiple or larger than a single lobe region, and the remaining 3 cases could not be clearly identified. The location and scope of leakage in the diagnosis of RT-3D-TEE were basically consistent with the intraoperative observation.RT-3D-TEE diagnosed 9 patients with partial flap ring dislocation, 6 patients with perivalvular abscess, and 3 patients with mechanical mitral valve sticking, which was consistent with the intraoperative observation. 2D-TTE diagnosed 6 patients with partial flap ring dislocation, 3 patients with perivalvular abscess and 3 patients with mechanical mitral valve sticking were misdiagnosed.There was no significant difference in the length and width of perivalvular leakage between the two methods (P > 0.05). The diagnostic accuracy of RT-3D-TEE was higher than that of 2D-TTE in the diagnosis of ring dislocation, perivalvular abscess and mechanical mitral valve valves.  Conclusion  RT-3D-TEE is a good complement to 2D-TTE in the diagnosis of perivalvular leakage after aortic valve replacement because of its superiority in the overall image of prosthetic valve because of its special observation position.
Effect of Different Surface Treatment Methods on Adhesion and Retention of Root Canal Wall

Abstract:
  Objective  To explore the effect of different treatment methods on adhesion and retention of root canal wall of fiber post .   Methods   50 single canal mandibular bicuspid teeth were collected and randomly divided into 5 groups. Group A was the control group; group B was treated with silane coupling agent; group C was the selective acid etching group; group D was treated with hydrogen peroxide solution, group E was the sandblasting treatment group. The mechanical test pieces were made according to the unified standard for mechanical test, and the surface appearance of the treated fiber pile was observed by scanning electron microscope.   Results  Co mparison of the maximum value and minimum value of tensile force in each group, the maximum values of group A, B, C, D and E were 13.16 MPa、13.42 MPa、18.31 MPa、 22.67 MPa and 21.05 MPa respectively, and the minimum values were 8.9 MPa, 8.94 MPa, 12.98 MPa, 17.69 MPa and 14.39 MPa respectively. The maximum and minimum values of samples in groups D and E were larger than those in group A, and were the largest among the five groups. Chi-square test was used to analyze the fracture modes of each experimental group, and the difference between the fracture modes was statistically significant (α < 0.05). Most of the fracture modes were the bonding interface damage between the adhesive and the fiber pile. The mixed damage in groups D and E was dominant, while the type I bond damage in groups A, B and C was dominant. Four different treatment methods all changed the surface apperance of the glass fiber pile to different degrees, the integrity of the fiber bundle was preserved, and the surface covering matrix was dissolved at different degrees.   Conclusion  A comprehensive evaluation of the four surface treatment methods of fiber post suggests that the fiber post treated with hydrogen oxide solution and sandblasting treatment is the most significant to improve the bond strength, and the fracture mode of the bond interface is mainly mixed bond failure, which is worthy of clinical application .

Abstract:
  Objective  To explore the application value of nutrition intervention combined with resistance exercise in senile sarcopenia, and to provide evidence for clinical intervention programs.   Methods  From August 2020 to May 2022, purposive sampling was used to select 50 patients with sarcopenia admitted to the Geriatrics Department of the Second People's Hospital of Kunming as the research subject. According to the order of the medical record number, they were divided into observation group and control group, with 25 cases in each group. The control group received routine diet, rehabilitation exercise and nursing, and the observation group received nutritional intervention combined with resistance exercise as well as the treatment of control group. Patients in both groups were treated for 2 months, and serum albumin (ALB), hemoglobin (Hb) and vitamin D (VD) levels were measured before and after intervention. Patient Generated-Subjective Global Assessment (PG-SGA) was used to evaluate the nutritional levels of patients before and after intervention. The modified Barthel index was used to evaluate daily living ability before and after intervention.   Results  Hb, ALB and VD in observation group were higher than those in control group, PG-SGA score was lower than that in control group, the difference was statistically significant (P < 0.05). The daily living ability of observation group was higher than that of control group, and the difference was statistically significant (P < 0.05).   Conclusion  Nutritional intervention combined with anti-resistance exercise can quickly restore the nutritional level of senile patients with sarcopenia and improve their daily living ability.

Abstract:
    Objective   To explore the epidemiological characteristics of Mycoplasma pneumoniae (MP) infection in children and the association between MP infection and meteorological elements.    Methods   Children admitted to the Second Affiliated Hospital of Kunming Medical University from January 2020 to December 2021 due to acute respiratory tract infection were retrospectively analyzed. Blood samples were collected for specific MP IgM and IgG double antibody titers, and environmental meteorological factors data were also collected. Descriptive statistics were used for clinical epidemiological characteristics of children with MP infection, logistic regression was used for correlation analysis, and regression Wald method was used to establish a relationship model.    Results   The positive rate of MP was 32.46% in 1260 test samples. The positive rates of MP in female and male children were 37.31% and 28.96%, respectively, and the difference was statistically significant (χ2 = 9.752, P < 0.05). The MP infection rate in infants was significantly lower than that in other age groups (χ2 = 67.68, P < 0.05). The detection rate in winter was significantly higher than that in other seasons (χ2 = 31.089, P < 0.05). Monthly cumulative precipitation (P = 0.001), PM2.5 (P < 0.001), and NO2P < 0.001) significantly increased the risk of MP infection.    Conclusion   MP infection in children in Kunming was significant associated with age, sex, season and meteorological evironment.
Simultaneous Imaging of Ureter and Abdominal Aorta and Its Application in Open Surgery for Abdominal Aortic Aneurysm

Abstract:
  Objective   To investigate the simultaneous ureter and abdominal aorta imaging technique, its clinical effect and application value in the open surgery of abdominal aortic aneurysm.   Method   From August 2021 to May 2023, 24 patients underwent open surgery in the Department of Cardiovascular Surgery, the Second Affiliated Hospital of Kunming Medical University, and the ureter and abdominal aorta were simultaneously imaged before surgery. Based on the adjustment of operating parameters and the results of the simultaneous development examination in these patients, the application of this technique in 24 cases of open surgery for abdominal aortic aneurysm was discussed, and the value of this technique for ureteral protection in open surgery for abdominal aortic aneurysm was expounded.   Result   During the simultaneous imaging of the ureter and abdominal aorta, CTA scans were performed 4-5 minutes after the first injection of 40 ml contrast agent. Most segments of the ureter were well displayed on CT and 3D images, and their contrast with surrounding tissues was relatively clear; There was no difference in the detection rate of ureteral abnormalities between simultaneous imaging of the ureter and abdominal aorta and intraoperative exploration (P > 0.05). There was no ureteral injury in 24 patients who underwent open surgery, and there was a statistical difference in the incidence of ureteral injury compared to the control group of 20 patients ( P < 0.05).   Conclusion   During the simultaneous imaging of the ureter and abdominal aorta, CTA scan performed 4-5 minutes after the first injection of 40 ml contrast agent showed good imaging of the ureter. It can clearly distinguish the ureter, abdominal aorta, and surrounding adhesive tissue, assist in ureteral localization, and its application in open surgery for abdominal aortic aneurysm can help to detect ureteral stenosis, peripheral adhesion, and other conditions before surgery, and avoid intraoperative ureteral injury.

Abstract:
  Objective  To investigate the clinical value of contrast-enhanced CT and MRI in preoperative TNM staging of pancreatic cancer.   Methods  A retrospective analysis was performed on 62 pancreatic cancer patients diagnosed by surgical pathology from January 2019 to December 2021 in the First Affiliated Hospital of Kunming Medical University. The preoperative enhanced CT and MRI images, operation status and postoperative pathological results were analyzed. All the findings were reviewed by two senior radiologists separately for preoperative imaging staging. The preoperative TNM staging of the two methods were compared with the surgical and pathological results.   Results  The overall accuracy of contrast-enhanced CT and MRI in TNM staging of pancreatic cancer were 86.96% (40/46)and 89.13% (41/46)in T stages, 78.57% (33/42)and 73.81% (31/42) in N stages, 96.77% (60/62) and 98.39% (61/62)in M stages, both had good consistency in T stages, moderate consistency in N stages and strong consistency in M stages compared with surgical and pathological results. There was no significant difference between the two methods in the accuracy of preoperative TNM staging (P > 0. 05).   Conclusion  Enhanced CT and MRI have a good correlation between preoperative TNM staging evaluation and surgical pathological results of pancreatic cancer, which can provide a reference for preoperative evaluation and guide treatment decision making for clinicians, and have good clinical application value.
Establishment and Evaluation of a Nomogram for Predicting Potential Death in Patients with Hepatitis C Cirrhosis and Sepsis in General Wards

Abstract:
  Objective   To explore the predictive factors of potential mortality in patients with hepatitis C cirrhosis and sepsis in general wards.   Methods  A retrospective analysis was performed on 194 patients with hepatitis C cirrhosis and sepsis diagnosed in the Third People’s Hospital of Kunming City from January 2012 to August 2022. Clinical data such as gender, age, comorbidity, site of infection, history of invasive operation within the last 2 weeks, MEWS score, length of stay, white blood cell/platelet, neutrophil count/lymphocyte count ratio (NLR), red blood cell and prothrombin time were collected. According to the prognosis, the patients were divided into two groups: death group (n = 13) and survival group (n = 181). The differences of various indicators between the two groups were analyzed and compared, and the influencing factors of death were analyzed.   Results  A total of 194 patients were included in the analysis, including 151 males (77.8%) and 43 females (22.2%). The average age was 50.6±9.7 years. There were 32 cases (16.5%) with hypertension and 32 cases (16.5%) with diabetes. Multivariate Logistic regression analysis showed that HCV infection time (OR = 1.114; 95%CI 1.000, 1.241), MEWS score (OR = 1.761; 95%CI 1.210, 2.564), no co-infection (OR = 0.081; 95%CI 0.010, 0.667), neutrophil/lymphocyte ratio (NLR) (OR = 1.084; 95%CI 1.021, 1.152) was an independent predictor of potential death in patients with hepatitis C cirrhosis and sepsis in the general ward. The sensitivity and specificity of the nomogram model are 92.3%, 81.2%, respectively; and the maximum Youden index is 0.735. The result of Hosmer-Lemeshaw test is χ2 = 2.471, P = 0.963 (P > 0.05), which indicates that the model fits well. ROC curve analysis showed that AUC = 0.916, 95%CI 0.856, 0.976. Calibration images of the Nomogram was used to verify the diagnostic value of the Nomogram model with an average absolute error of 0.022. The results showed that the actual and predicted values of bacterial infection were in good consistency.   Conclusion  Time of HCV infection, MEWS score, unco-infected, and NLR were associated with the risk of death from hepatitis C cirrhosis and sepsis. MEWS combined diagnosis can improve the accuracy of predicting death in patients with hepatitis C cirrhosis and sepsis in general wards.

Abstract:
  Objective  To investigate the therapeutic efficacy and safety of bortezomib and lenalidomide combined with dexamethasone in patients with Multiple Myeloma (MM).   Methods  A total of 94 patients with multiple myeloma in the Hematology Department of the First People’s Hospital of Qujing City from September 2021 to September 2022 were selected and divided into 2 groups by random number table method. The control group (n = 47 cases) was treated with bortezomib combined with dexamethasone, and the observation group (n = 47 cases) bortezomib, dexamethasone and lenalidomide. One course of treatment lasted for 3 weeks, and a total of 6 courses of treatment were conducted. The effective rate, blood biochemical indexes, immune level, disease load and adverse reactions of the two groups were compared.   Results  After treatment, the effective rate of observation group was higher than that of control group, the difference was statistically significant (P < 0.05). After treatment, bone marrow plasma cells and blood M protein levels in the observation group were lower than those in the control group, and the difference was statistically significant (P < 0.05). BAP, OPG and LDH in observation group were higher than those in control group, and the differences were statistically significant (P < 0.05). CD3+, CD4+, CD4+/CD8+ in observation group were higher than those in control group, and CD8+ were lower than those in control group, the difference was statistically significant (P < 0.05). The percentage of bone marrow plasma cells, the level of M protein and the level of free light chain were decreased in 2 groups. There was no significant difference in the incidence of bone marrow suppression, fingertip numbness, alopecia, kidney damage and gastrointestinal reaction between 2 groups (P > 0.05).   Conclusion  Bortezomib, lenalidomide combined with dexamethasone for 6 consecutive courses of treatment can improve the treatment efficiency of multiple myeloma patients, improve their blood biochemical indexes, increase the levels of CD3+, CD4+, CD4+/CD8+, and significantly improve the drug safety factor, worthy of promotion and application.

  Objective   To study the genetic polymorphism of 17 autosomal STR loci in the Jino people of Yunnan, and to evaluate its value in forensic medicine.   Methods   A total of 331 unrelated individuals of the Jino people in Yunnan Province were examined. AGCU 17+1 kit was used for STR genotyping, and ABI3130 capillary electrophoresis apparatus was used to isolate the amplified product fragments and to determineallele type .   Results   A total of 186 alleles were detected, and the allele frequencies ranged from 0.0015 to 0.561 9. TPOX and TH01 loci had the lowest polymorphism and recognition ability, and D3S1358, D13S317, D16S539, CSF1PO and D5S818 had poor polymorphism parameters. Other loci showed better polymorphism. The distribution of genotypes was in accordance with Hardy-Weinberg equilibrium, and CDP and CPE showed good recognition.   Conclusion   Except for TPOX and TH01, the remaining 15 autosomal STR loci have good polymorphism in Jino population, which can meet the application of forensic individual identification and paternity identification. Except the TPOX and TH01 loci, the remaining 15 autosomal STR loci have good polymorphisms in the Jino population.
Application of IBS and FSI Methods in the Full Sibling Testing of Yunnan Population

Abstract:
  Objective  To explore the applicability of IBS and FSI methods in the identification of full sibling relationship in Yunnan population.   Methods  A total of 448 pairs of full siblings and 1020 pairs of unrelated individuals were genotyped for 19 STR loci, and CIBS and CFSI were calculated. Descriptive statistics, normality test and t-test were performed for lgCFSI values and CIBS values of whole sibling pairs and unrelated individual pairs using SPSS 25.0 software. χ2 test was used for counting data.  Results  The lgCFSI and CIBS values of the whole sibling group and the unrelated individual group were in normal distribution. The difference between the unrelated individual group and the full sibling group was statistically significant in both the IBS and FSI methods. The statistical comparison of the difference between the three cutt-offs dividing methods in the old and new standards is statistically significant. For 19 autosomal STR loci, the system power of the three methods in the two standards were 0.7500, 0.5565 and 0.6625, respectively, while the system power of the Yunnan population in this study were 0.7405, 0.5109 and 0.6621, respectively.  Conclusion  The new standard has a high value of forensic medicine application in the identification of full sibling relationship in Yunnan. The IBS method and FSI method based on 19 autosomal STR loci detection system can effectively reduce the risk of misjudgment in the identification of full sibling relationship.
Correlation between DNA Detection Rate and Sweat Pore Density of Palm Touch Biological Samples

Abstract:
  Objective  To study the relationship between the detection rate of touch DNA and sweat pore density in male palm touch biological samples, and explore the factors affecting the detection rate of touch DNA.   Methods  The right hand hypothenar palmprints of 73 male college students were collected by using the added layer fingerprinting method, and the sweat pore count was performed in specific site . Palm-touch biological samples of 35 individuals were collected by direct palm wiping method and indirect glass slide transfer wiping method. PCR was performed on the samples by direct amplification, and short tandem repeat (STR) results were analyzed. The correlation between STR allele detection rate and sweat pore density was analyzed.   Results  The success rate of comprehensive sweat pore collection was 84.93%, the density of the sweat pore was (531.41±97.69)/cm2, the minimum value was 309 /cm², and the maximum value was 814 /cm2. There was no correlation between the palm sweat pore density and the STR allele detection rates using direct and indirect wiping methods (P1 = 0.806, P2 = 0.701).   Conclusion   Two different experimental methods were used to test the DNA of the palm touch biological samples, both of which showed that there was no correlation between the sweat pore density and the detection rate of touch DNA, indicating that the sweat pore density was not the main or only influencing factor of the detection rate of touch DNA.

Abstract:
Early-onset scoliosis (EOS) is a three-dimensional spinal deformity in children. At present, the main clinical treatment methods include follow-up, surgical treatment and non-surgical treatment, while non-surgical treatment can delay and control the progression of the disease and buy time for growth and development as well as surgical treatment. Serial casting, traction therapy and targeted exercise for scoliosis are all non-surgical treatments that have been used in clinical practice for many years. With the progress of technology and treatment, new ideas have been provided for the non-surgical treatment of EOS, such as the change of treatment mode and site of action, pharmacological research, mental health monitoring, etc. This article summarizes the nonsurgical treatment methods and new ideas for the development of early onset scoliosis.

Abstract:
Diffuse large B-cell lymphoma is the most common type of non-Hodgkin lymphoma. At present, CD20 monoclonal antibody combined with chemotherapy has increased the response rate. However, some people still have poor efficacy or even drug resistance, and new drugs are still needed to improve the response rate. Regulatory T cells play an important role in immune regulation via coordinating cellular and molecular networks in tumor microenvironment in diffuse large B-cell lymphoma, and mediating local immunosuppression, consequently promoting tumor cell proliferation and migration. Therefore, it is promising to target the treatment of diffuse large B-cell lymphoma. This article reviews the mechanisms of regulatory T cells in the diffuse large B-cell lymphoma microenvironment.

Abstract:
Oral squamous cell carcinoma (OSCC) is the most common malignant tumor in the oral and maxillofacial region with high incidence and poor prognosis. Cancer cells change their metabolism through glycolytic metabolic reprogramming to support their increased demand for ATP. Glycolytic reprogramming mediates the development of multisystem tumors, including oral squamous cell carcinoma, involving multiple signaling pathways and key factors. In this paper, the changes of glycolytic metabolism, key factors and potential clinical diagnostic and theraputic approches of OSCC are summarized, and the role of OSCC in oral squamous cell carcinoma is reviewed in order to provide new research directions and ideas.

Abstract:
  Objective  To improve pulmonary rehabilitation compliance and health literacy in patients with chronic obstructive pulmonary disease.   Methods  A total of 96 COPD patients were selected as research subjects and divided into experimental group and control group by random number table method, with 48 cases in each group. The control group received routine nursing, health education and family pulmonary rehabilitation guidance, and the experimental group received pulmonary rehabilitation intervention guided by BCW theory, including breathing exercise, muscle relaxation exercise, breathing exercise exercise, walking and marching exercise. Pulmonary rehabilitation compliance, health literacy levels, improved Medical Research Council Respiratory Questionnaire (mMRC) scores, COPD Assessment Test (CAT) scores and pulmonary function indicators were compared between the two groups at 6 months after intervention.   Results  After intervention, the proportion of complete compliance of pulmonary rehabilitation compliance in the experimental group (60.4%) was better than that in the control group (25%). The level of health literacy, mMRC score, CAT score and pulmonary function index of the two groups were improved, and the difference between the two groups was statistically significant (P < 0.05).   Conclusion  The BCW-guided pulmonary rehabilitation intervention program can effectively improve the compliance of pulmonary rehabilitation and health literacy of patients with COPD, improve the lung function and quality of life of patients.

  Objective  To explore the application effect of “structure-process-outcome” three-dimensional quality evaluation model in the prevention and treatment of surgical inpatients with VTE.   Methods  A total of 400 patients admitted to Department of General Surgery in Yan’an Hospital of Kunming City from July 2020 to June 2021 were selected as research subjects, and were divided into control group and experimental group using random number table method, with 200 patients in each group. The control group received hospital VTE prevention and treatment nursing following the hospital regulations. The experimental group received the “structure-process-result” three-dimensional quality evaluation model, which was applied to the prevention and management system of VTE in the hospital, and was optimized from three dimensions: structure, process and result. The incidence of VTE, patient satisfaction, patient service perception and nursing quality score of the two groups were compared.   Results  The incidence of VTE in the experimental group was significantly lower than that in the control group (χ2 = 4.592, P < 0.05). There were significant differences in patient satisfaction (t = -8.811.91, P < 0.001) and service perception (t = -11.813, P < 0.001) between experimental group and control group. There were differences in five dimensions of service perception between the experimental group and the control group, namely, service system (t = -7.401, P < 0.001), nursing professionalism (t = -8.347, P < 0.001), executive ability (t = -5.73, P < 0.001), demand feedback (t = -8.024, P < 0.001), nurse-patient relationship (t = -9.799, P < 0.001). The experimental group scored significantly higher on patient satisfaction and service perception than the control group. In addition, the nursing quality of the experimental group showed statistically significant differences, SD scores of nursing quality gradually decreased, and nursing quality gradually stabilized.   Conclusion  The application of the “structure-process-result” three-dimensional quality evaluation model can reduce the incidence of VTE in surgical inpatients and promote the high-quality development of nursing teams.
Effect of HAPA Qualitiative Exercise Management on Exercise Compliance of Patients with Coronary Heart Disease Based on Excellent Care Platform

Abstract:
    Objective   Help stable patients with coronary heart disease effectively improve exercise compliance after discharge, and achieve self-health management for patients with coronary heart disease.    Methods   70 patients with stable coronary heart disease were divided into control group (35 cases) and intervention group (35 cases) by random number table method. The control group received cardiac rehabilitation nursing, while the intervention group received a “HAPA quantitative exercise management model based on excellent care platform” in cardiac rehabilitation nursing care. The effect of "HAPA quantified exercise management mode based on excellent care platform on exercise compliance of patients in 2 groups was compared and analyzed.    Results   After 12 weeks of intervention, the exercise frequency in the intervention group was significantly higher than that in the control group (P < 0.05). After 12 weeks of intervention, the exercise time in the intervention group was longer than that in the control group, with a statistically significant difference (P < 0.05). Repeated measurement ANOVA was performed on the weekly exercise time of the two groups, and the time effect, inter-group effect and interaction effect were statistically significant (P < 0.01). At 6 weeks and 12 weeks after intervention, the weekly exercise duration in the intervention group was better than that in the control group, with statistical significance (P < 0.05). The duration of exercise per week in the intervention group was better than that in the control group as time progressed.    Conclusion   HAPA quantitative exercise management based on the excellence care platform" can effectively improve the exercise compliance of patients with stable coronary heart disease. The quantified FITT exercise management solution fits patients’ exercise preferences while promoting safe, regular exercise. According to different behavioral stages of patients, HAPA implements corresponding quantitative exercise management measures to promote the transformation of patients’ motor behavior intentions into external behaviors. The use the excellent care platform to supervise and guide the quantitative exercise management of HAPA effectively guarantees the generation and maintenance of exercise behavior in patients with coronary heart disease.
