Comparative Studies on Different Methods of Collecting Human Sloughed Epithelial Cells from Various Carriers

  • Objective To comparatively study different methods of collecting human sloughed epithelial cells from various carriers.Methods The used face masks, clothes and gloves were selected.The sloughed epithelial cells were collected from the materials using the following different methods:adsorption by negative pressure, sticking with double-sided adhesive, scissoring and wiping.DNA was exacted using QIAGEN QIAamp DNA Investigator Kit.PCR was conducted using AB Identifiler kit.PCR product was separated by AB 3130 XL.STR typing was performed by Genemapper ID v3.2 software.Results To collect human sloughed epithelial cells from small carriers such as face masks and gloves, all the methods (adsorption by negative pressure, sticking with double-sided adhesive, scissoring and wiping) were useful.When big carriers such as clothing were mentioned, both methods (adsorption by negative pressure and scissoring) were useful.Conclusions For permeable carriers such as textile products, the method of adsorption by negative pressure can be used to collecting human sloughed epithelial cells from big carriers.The method of scissoring directly can be used to collecting human sloughed epithelial cells from carriers with clear spots.The method of sticking with double-sided adhesive can be used to collecting human sloughed epithelial cells from small carriers.
