Overexpression of miR-4328 Inhibits the Proliferation, Migration and Epithelial Mesenchymal Transition of Prostate Cancer Cells

  • Objective To study the role and molecular mechanism of miR-4328 in prostate cancer.Methods CCK-8 method was used to detect the proliferation of prostate cancer cells, and Transwell method was used to detect the migration of prostate cancer cells, and Real-time PCR and Western blotting method were used to detect the epithelial mesenchymal transition markers and the expression of ZEB1.Results The expression level of miR-4328 in prostate cancer tissues was significantly lower than that in paracancerous tissues.Overexpression of miR-4328 in prostate cancer cells significantly inhibited the cell proliferation (P<0.001) .And overexpression of miR-4328 also significantly inhibited the migration of prostate cancer cells (P<0.001) .And overexpression of miR-4328 up-regulated the epithelial cell marker E-cadherin, and down-regulated the mesenchymal cell markers including N-cadherin and Vimentin (P <0.001) transition of prostate cancer cells.Further studies found that overexpression of miR-4328 could down regulate the expression level of epithelial mesenchymal transition related transcription factor ZEB1 (P<0.001) .Conclusion Overexpression of miR-4328 can inhibit the proliferation, migration and epithelial mesenchymal transition of prostate cancer cells.
