The Effect of GSPs-TACE on the Growth of Liver Transplant Tumors in Rabbits

  •   Objective  To investigate the effect of gelatin sponge particle chemotherapy combined with hepatic artery chemoembolization on the growth of rabbit liver transplant tumors.   Method  Sixty New Zealand white rabbits were randomly divided into a model control group and gelatin sponge particles (GSPs)- Transcatheterarterial chemoembolization. The TACE group, the epirubicin-TACE group and the GSPs+ epirubicin-TACE group (n = 15). VX2 cell liver cancer models were established in all experimental rabbits. After the operation, in the GSPS-TACE group, GSPs were perfused during TACE. In the epirubicin-TACE group, epirubicin solution (1mg/mL) was perfused. In the GSPs+ epirubicin-TACE group, a mixture of epirubicin solution and GSPs was used for embolization. Computed tomography was used to detect the tumor volume of experimental rabbits in each group. Hematoxylin-eosin staining was used to observe the pathological morphology. The apoptosis rate of tumor cells was detected by Terminal Deoxynucleotidyl Transferase-Mediated dUTPNick End Labeling (TUNEL) staining; Immunohistochemical detection of positive expression of vascular endothelial growth factor (VEGF); Western blotting was used to detect B-cell lymphoma 2 (Bcl-2), Bcl2-associated X protein (Bax), and cysteine-aspartic acid Protein expression of protease-3, caspase-3.   Results  Compared with the model control group, the tumor volume, the proportion of intrahepatic metastasis, the proportion of intra-abdominal metastasis, VEGF, and Bcl-2 in the GSPs-TACE group were all decreased (P < 0.05), while the apoptosis rate of tumor cells, Bax, Bcl-2/Bax, and caspase-3 were all increased (P < 0.05). There was no statistically significant difference between the GSPS-TACE group and the epirubicin-TACE group (P > 0.05). Compared with the epirubicin-TACE group, the tumor volume, the proportion of intrahepatic metastasis, the proportion of intra-abdominal metastasis, VEGF, and Bcl-2 in the GSPs+ epirubicin-TACE group were all decreased (P < 0.05). The apoptosis rate of tumor cells, Bax, Bcl-2/Bax, and caspase-3 all increased (P < 0.05).   Conclusion  Gelatin sponge microparticle chemotherapy combined with hepatic artery chemoembolization inhibits the growth of rabbit liver transplant tumors.
