Effects of Different Penetration Enhancers on in vitro Transdermal Permeation of Bagui Zhitong Cream

  •   Objective  To study the best preparation method of Bagui Zhitong cream, to determine the best matrix formulation, and to investigate the effect of different Penetration Enhancers on in vitro Transdermal Permeation of Bagui Zhitong Cream.   Methods  We prepared Bagui Zhitong Cream with different concentrations of penetration enhancer of azone. Modified Franz diffusion cell was adopted using mouse skin of isolated male rat as transdermal barrier. The content of Cinnamaldehyde was determined by UPLC. The accumulative transdermal amount (Q)and percutaneous permeability (J)were calculated and compared between different concentrations of BaGui Zhitong cream to select the best concentration of Penetration Enhancers.   Results   Different concentrations of azone promoted Cinnamaldehyde in vitro permeation in different degrees, the order of promoting infiltration was as follows: 5% azone > 3% azone > 7% azone > 1% azone > 10% azone. Through the multiple comparison, the difference had statistical significance (P < 0.05).   Conclusion  In Bagui Zhitong cream containing 1% azone, 3% azone, 5% azone, 7% azone and 10% azone, 5% azone as penetration enhancer has the best effect in Bagui Zhitong Cream.
